摘    要:目的观察特异性DNA甲基转移酶抑制剂5-氮-2’-脱氧胞苷(5-Aza-CdR)对人脑星形细胞瘤细胞侵袭力的影响,并探讨其机制。方法分别将终浓度为0、1×10^-7、5×10^-7、1×10^-6mol/L5-Aza-CdR加入星形细胞瘤细胞作用48h,采用Boyden小室观察星形细胞瘤细胞的侵袭力,采用明胶酶谱分析法检测星形细胞瘤细胞基质金属蛋白酶9(MMP-9)的表达变化。结果随着5-Aza-CdR浓度的升高,星形细胞瘤细胞穿膜细胞数呈升高趋势,proMMP-9及MMP-9的表达呈增高趋势(P均〈0.05)。结论5-Aza—CdR可增强人脑星形细胞瘤细胞侵袭力,可能与DNA低甲基化上调MMP-9表达有关。

Abstract:Objective To investigate the effect of 5-Aza-Z'-deoxycytidine (5-Aza-CdR) on invasion of astrocytoma cells, and explore its mechanism. Methods Astrocytoma cells were treated with different concentrations of 5-Aza-CdR for 48 h. The invasion ability of astrocytoma ceils and the expression of MMP-9 were detected by Boyden chamber experiment and gelatin zymography. Results Along with the increase of the concentration of 5-Aza-CdR, the number of cells traversed through the membranes was on the rise; the expression of proMMP-9 and MMP-9 raised up gradually (P 〈 0.05 ). Conclusion 5-Aza-CdR can enhance the invasion ability of astrocytoma cells, and it may be related to the upregulation of the expression of MMP-9 with DNA hypomethylation.
