p27^kip1、cyclin D1、cyclin D3及Skp2在肾细胞癌中的表达及临床意义

摘    要:目的:检测p27^kip1、cyclin D1、cyclin D3及Skp2在肾细胞癌组织和正常肾组织中的表达情况,探讨几种蛋白之间的相关性及其与肾细胞癌各项临床病理参数的关系。方法:采用组织微阵列技术及免疫组化SP法检测p27^kip1、cyclin D1、cyclin D3及Skp2在80例肾细胞癌及40例正常肾组织中的表达并作统计学分析。结果:①肾细胞癌中p27^kip1阳性表达率低于正常肾组织(P=0.007),cyclin D1、cyclin D3、Skp2阳性表达率高于正常肾组织(分别P=0.001,P=0.04,P=0.024);②p27^kip1与Skp2表达强度呈负相关(r=-0.262,P=0.019),p27^kip1与cyclin D1表达强度呈负相关(r=-0.285,P=0.010),cyclin D1与cyclin D3之间呈正相关(r=0.314,P=0.002);③p27^kip1的表达与肿瘤分化程度及临床分期呈负相关(P〈0.05),Skp2表达与肿瘤分化程度呈正相关(P=0.002)。结论:Skp2、p27^kip1的表达与肾细胞癌发生及生物学特性密切相关,为肾细胞癌诊治的研究提供了新的思路和依据。

Abstract:Objective:To investigate the expression of p27^kip1,cyclin D1, cyclin D3 and skp2 in renal cell carcinoma(RCC) and normal renal tissues. According to the cases' clinical and pathological data, the relationship between the protein and the clinical or pathological parameters and the inherent connections of the proteins were discussed, in order to obtain the meaningful judgement factor to prognosis. Methyls:Tissue microarrays of clinical samples were stained using immunohistochemical way to observe the expression and location of p27^kip1 , cyclin Dl,cyclin D3 and skp2 protein. Analyses were carried out using SPSS 13.0 software. Results: ①Low expression of p27^kip1 was found in (43.8 % ) renal cell carcinoma. Expression of cyclin D1,D3 and Skp2 in renal cell carcinoma was found significantly higher than that in normal samples;②The expression of p27^kip1 was inversely correlated with the expression of Skp2( r = 0. 262. P =0. 019) and cyclin D1 ( r =--0. 285, P =0. 010). The expression of cyclin D1 was strongly correlated wilh the cxpres sion of cyclin D3 ( r =0. 314, P =0. 002); ③Inverse correlations were presenl between p27^kip1 expression and tumor grade,or clinical stage( P 〈0.05). A direct correlation was present belween Skp2 expression and tumor grade ( P =0. 002). Conclusion:The expression of Skp2 and p27^kip1 showed close correlation with the development and clinical biological characteristics of RCC, which could be a new clue for the research of RCC.
