脂联素受体2在糖尿病大鼠肝脏中的表达

目的探讨脂联素受体2在1型和2型糖尿病大鼠肝脏中的表达差异以及与糖尿病发病的可能机制的关系。方法 30只SD大鼠分为3组：对照组（con）,腹腔注射等体积量枸橼酸缓冲液;1型糖尿病组（1-DM）：一次性腹腔内注射高剂量链脲佐菌素60 mg/kg;2型糖尿病组（2-DM）：大鼠高脂饲料喂养1个月,禁食12 h后腹腔注射低剂量链脲佐菌素35 mg/kg,继续高脂饮食喂养。采用ELISA试剂盒检测血清胰岛素和脂联素水平,免疫组化法分析肝脏脂联素受体2（Adipo R2）的表达,HE染色观察各组肝脏病理改变。结果糖尿病组大鼠血糖均高于对照组,与con组（8239±2259）ng/ml比较,1-DM组血清脂联素降低（5266±1985）ng/ml,P〈0.05],肝Adipo R2表达无明显改变（13.39±3.09）%,（11.92±5.62）%,P﹥0.05];与con组（8239±2259）ng/ml和1-DM组（5266±1985）ng/ml比较,2-DM组血清脂联素（3533±1048）ng/ml降低（P〈0.05）,肝Adipo R2表达升高（69.76±10.8）%,P〈0.01,且有脂肪变性等损伤。结论肝Adipo R2在1-DM大鼠肝脏的表达与正常组无异,在2-DM大鼠肝脏高表达,与2-DM肝脏的损伤和脂代谢异常相关。
