海参皂苷抑制人脑胶质母细胞瘤U251细胞生长实验研究

摘    要:目的研究一种新型海参皂苷fuscocineroside A对人脑胶质母细胞瘤U251细胞的生长抑制作用及其机制。方法采用四甲基偶氮唑蓝(MTT)法绘制细胞生长曲线;用Hoechst33342荧光染色、末端脱氧核苷酸转移酶介导的生物素脱氧尿嘧啶核苷酸缺口末端标记法(TUNEL)及流式细胞仪检测分析U251细胞凋亡;并通过Western blot检测细胞内survivin蛋白表达情况。结果Fuscocineroside A能显著抑制U251细胞的生长,诱导其凋亡,下调survivin的表达,呈浓度及时间依赖性,而对正常胶质细胞并没有明显抑制作用。结论Fuscocineroside A能诱导U251细胞凋亡,这种作用可能与survivin的表达下调有关。

Abstract:Objective To explore the potential inhibitive effect of fuscocineroside A on proliferation of human glioblastoma U251 cells and its mechanism.Methods Methyl thiazolyl tetrazolium(MTT)assay was performed to measure the proliferation of U251 cells and normal astrocytes with different concentrations of fuscocineroside A.Hoechst33342 fluorochrome stain,terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick end labeling(TUNEL)and flow cytometry were used to observe the morphologic changes and the apoptosis.Western blotting was performed to detect the expression of survivin in U251 cells.Results Fuscocineroside A could significantly inhibit the proliferation and induce the apoptosis and down-regulate the survivin expression in U251 cells in a concentration and time-dependent manner,but it could exhibit slight cytotoxicity to normal human astrocytes.Conclusion Fuscocineroside A can induce apoptotic death in U251 cells,which may be related to the downregulation of survivin expression.
