丝光绿蝇针刺诱导血淋巴特性研究

摘    要:目的探讨针刺体壁诱导后丝光绿蝇血淋巴的部分性质.方法丝光绿蝇Ⅲ龄幼虫针刺体壁处理后48h提取血淋巴,观察其对大肠埃希菌、金黄葡萄球菌、白色葡萄球菌、变形杆菌、伤寒杆菌、副伤寒杆菌、枯草杆菌、溶壁微球菌的抑菌效果.设置3μl、5 μl、7 μl不同的血淋巴加样量实验组,记录不同加样量对金黄葡萄球菌的的抑菌效果;设置不同稀释浓度,记录不同浓度的血淋巴对金黄葡萄球菌的抑菌效果.诱导后48h的血淋巴置于冰箱保存后,设置36℃和100℃不同解冻温度,观察不同温度解冻的血淋巴对金黄葡萄球菌和大肠埃希菌的抑菌效果.诱导后48h的血淋巴,100℃水浴3 min,观察其对金黄葡萄球菌的抑菌效果.结果诱导后48h的血淋巴,1/2、1/4、1/8、1/16、1/32、1/64的不同稀释浓度,对金黄葡萄球菌均能产生明显的抑菌环.丝光绿蝇诱导后48h的血淋巴,对上述8种细菌都能产生明显的抑菌环,对溶壁微球菌抑菌环最大,对金黄葡萄球菌和枯草杆菌的抑菌环次之.不同的加样量实验,以加7μl的抑菌效果最佳;36℃和100℃不同温度解冻,对金黄葡萄球菌和大肠埃希菌均有抑菌效果,36℃解冻效果好于100℃;100℃水浴3min,对金黄葡萄球菌仍有抑菌活力.结论丝光绿蝇针刺体壁诱导后48 h的血淋巴,具有较强和较广的抗菌活力,抗菌活力具有一定的热稳定性,抑菌试验加样以7μl为宜,解冻以36℃为宜.

Abstract:Objective To investigate the characters of the hemolymph induced by pricking on larvae of Lucilia sericata.Methods After being induced by pricking, the hemolymph was collected at 48 h, and tested against eight species of medical bacteria respectively. Diameters of clear inhibitory ring were observed and recorded to express antibacterial activity. The immunized hemolymph was also used in the different dosage assay, thawing assay at different temperatures, antibacterial assay after treatment at high temperature, antibacterial assay at different concentrations. Results It was found that the antibacterial effect of 7 μl dosage group was better than the others. The effect of thawing at 36℃ was better than at 100℃. The immunized hemolymph possessed antibacterial activity against eight species of medical bacteria such as Staphylococcus aureus, Staphylococcus albus, Escherichia coli, Proteus vulgaris, Salmonella typhi, Salmonella paratyphi, Bacillus subtilis, and Micrococus lysodeikticus. The immunized hemolymph also possessed antibacterial activity against S. aureus after bathed at 100℃ for 3 min, and against S. aureus after diluted by 64 times. Conclusion It implies that, the dosage of 7 μl and the thawing temperature of 36℃ should be chosen in the assay. It is also suggested that the immunized hemolymph collected at the 48 h after treatment should possess wide antibacterial spectra and should keep stable antibacterial activity after treatment at high temperature.
