摘    要:目的探讨脑内局部给药建立颞叶癫痫(EP)模型方法及其行为与病理学改变。方法通过立体定向术向大鼠海马局部注射海人酸(KA)而建立颞叶EP模型,观察其行为学、脑电图和病理学改变情况。结果KA注入海马后大鼠表现出凝视、湿狗样抖动、口的咀嚼运动、点头、肢体阵挛等表现。随着时间的推移,大鼠主要表现为阵发性旋转,并身体立起、向上窜跳、跌倒、四肢抽搐,约10h后发作停止,逐渐恢复到正常大鼠状态,以后每周发作1~2次,主要为Ⅱ~Ⅳ级。结论大鼠脑内局部注入KA后EP发作明显,其行为症状、海马病理改变类似人类颞叶EP,可作为很好的研究颥叶EP的工具。

Abstract:Objective To explore a better method to establish an animal model of temporal epilepsy in rats and observe the changes of its behavior and pathology.Methods Kainic acid(KA) with the dosage calculated according to the weight was injected into rat's hippocampus by stereotactic operation.Results After the rats awoke from anesthesia,wet-dog shakes,lips and head self-movement,convulsion of forelimbs and hind limbs occurred successively.The seizures were paroxysmal with increased intervals, especially rotation seizures were manifest.The rats' behavior gradually recovered to normal after 10 hours.Then the seizure occurred 1-3 times every week.Conclusion The seizure pattern and pathological basis of temporal epilepsy in the rat model established by stereotactic management were almost the same as those in human beings.The kainic acid induced model is an ideal animal model simulating human temporal lobe epilepsy.
