Abstract:Objective To investigate the isolation and identification of curcumol and its anti-tumor mechanism in vitro.Methods Zedoary turmeric oil was extracted by vacuum distillation and recrystallized to obtain curcumol,which was identified by oleyl ether-ethyl acetate strip.Cervical cancer cells and lung cancer cells were cultured in vitro,and their proliferation was detected by MTT.The cells were divided into model group,curcumol group and Zedoary turmeric oil group.The apoptosis rate was detected by flow cytometry,and the expression of Ki67 protein and PCNA protein in tumor cells was detected by Western blot.Results Curcumol could be isolated by vacuum distillation,and its purity was 89.57%.Both curcumol and Zedoary turmeric oil could effectively inhibit the proliferation of cervical cancer cells and lung cancer cells,and the inhibition rate of curcumol on cervical cancer cells and lung cancer cells is higher than that of Zedoary turmeric oil.Compared with those in the model group,the apoptosis rate and the expression levels of Caspase-3 and Bax protein in curcumol group and Zedoary turmeric oil group were significantly increased,while the expression levels of Ki67,PCNA and Bcl-2 protein in curcumol group and Zedoary turmeric oil group were significantly decreased(P<0.05).Compared with those in Zedoary turmeric oil group,the apoptosis rate and the expression levels of Caspase-3 and Bax protein in curcumol group were significantly increased,while the expression levels of Ki67,PCNA and Bcl-2 protein in curcumol group were significantly decreased(P<0.05).Conclusion Curcumol isolated from Zedoary turmeric oil by vacuum distillation can effectively inhibit the proliferation of tumor cells and promote their apoptosis,and the effect of curcumol is significantly better than that of Zedoary turmeric oil.
