奥硝唑缓释膜体内、外释放性的研究

摘    要:目的:测定奥硝唑缓释膜体内、外释放度。方法:体外释放度试验采用《中国药典》“转篮法”;体内释放度实验取大鼠于牙周袋内置入缓释膜(0.18mg),给药后不同时间测定龈沟液中奥硝唑浓度,并与大鼠腹腔注射奥硝唑溶液(25mg)比较。结果:缓释膜在体外试验中10min释药13.9%,10h后仍有药物释放;腹腔注射给药后龈沟液药物浓度是牙周袋内局部置药的1%~5%。结论:该缓释膜具有良好的缓释功能,能在大鼠牙周袋内较长时间维持有效浓度。

Abstract:OBJECTIVE:To determine the release rates of ornidazole slow-release membranes in vitro & in vivo. METHODS:The release rate of ornidazole slow-release membranes in vitro were determined by the rotating basket method adopted from Chinese Pharmacopeia. The release rate test in vivo was performed by inlaying periodontal pocket of rats with slow-release membranes(at a dosage of 0.18mg),the level of ornidazole in gingival crevicular fluid was determined at different time after administration, and the determination results were compared with that in rats receiving intraperitoneal administration of ornidazole(at a dosage of 25mg). RESULTS:The release rate of the slow-release membrane in vitro was 13.9% in the initial 10 minutes, and drug release could also be noted 10 hours later. The level of ornidazole in gingival crevicular fluid in rats receiving intraperitoneal injection was 1%~5% of that in rats receiving local administration in periodontal pocket. CONCLUSION:The ornidazole slow-release membrane shows satisfactory slow-release property and it can maintain at a valid concentration for a long time in periodontal pocket of rats.
