心肌亚细胞Ca^2+分布电镜细胞化学定位研究

摘    要:应用Ca ̄(2+)细胞化学探针—焦锑酸钾技术,结合电镜X线电子探针显微分析技术,研究心肌细胞内Ca ̄(2+)定位、分布。结果:正常心肌细胞Ca ̄(2+)以细小颗粒形态主要位于肌浆网腔、肌膜、细胞核,而胞浆和线粒体Ca ̄(2+)分布少。X线电子探针证实沉淀颗粒主要为Ca ̄(2+),直接证实心肌亚细胞Ca ̄(2+)的高度隔空化分布,为进一步深入研究心肌缺血再灌注等Ca ̄(2+)异常代谢变化提供新的研究方法。

Abstract:pplying Ca2 cytochemistry probe-putassium pyroantimonate(PPA)and electron probe micro-analysis(EPMA)techniques,Westudied the localization and distribution of calcium in myocardial sub-cell.The results demonstrated that calcium was mainly localization within sarcoplasniic reticulum, sar-colemma and nucleus,and calcium precipitates was less in cytoplasm and mitochOndria in normal my-ocardium. X-ray electron probe confirmed directly that Ca2 was the main component in precipitate par-ticles and high compartmental distribution. PPA combined witb EPMA technique provided new experi-mental method to the study of Ca2 paradox in myocardial ischemia and reperfusion etc.
