DNA甲基转移酶1与CD11a基因在系统性红斑狼疮患者中mRNA水平的表达

摘    要: 目的 检测系统性红斑狼疮(systemic lupus erythematosus,SLE)患者中DNA甲基转移酶1(DNA methyltransferase 1,DNMT1)和CD11a基因的表达水平,探讨DNMT1的表达异常在SLE发病中的作用。方法 用Real-time PCR方法测定SLE患者缓解期、活动期和正常人外周血单个核细胞(peripheral blood mononuclear cell,PBMC)中DNMT1和CD11a基因的表达水平。结果 SLE活动期患者PBMC中DNMT1的表达水平较对照组(P=0.014)和缓解期组(P=0.030)显著下降;缓解期的DNMT1表达水平与对照组(P=0.264)相比差异无统计学意义;SLE患者DNMT1的表达水平与SLE疾病活动指数(SLE disease activity index,SLEDAI)呈现负相关(P<0.05)。SLE活动期患者PBMC中CD11a的表达水平较对照组(P=0.003)显著升高(P<0.05);缓解期的CD11a表达水平与对照组(P=0.250)和缓解期组(P=0.069)相比差异无统计学意义(P>0.05);SLE患者CD11a的表达水平与SLEDAI呈正相关(P<0.05)。SLE患者DNMT1与CD11a的表达水平无显著相关性(P>0.05)。结论 SLE患者PBMC中DNMT1表达水平的降低可能是造成基因组DNA低甲基化的重要原因,使得CD11a等致病基因过度表达,进而导致SLE发病。
