双氟甲基鸟氨酸抑制大鼠脑缺血后迟发性神经元死亡的研究

摘    要:观察阻断脑血流10min后腹腔注射双氟甲基鸟氨酸(DFMO)大鼠海马CA1区域迟发性神经元死亡(DND)现象的变化。结果显示:DFMO对大鼠海马CA1区神经元有明显的保护作用,并且在一定范围内与DFMO的剂量有正相关关系。DFMO的上述作用只有在脑缺血后及时给药才能出现。实验还提示:多胺及其代谢限速酶鸟氨酸脱羧酶(ODC)在DND的产生中具有一定作用。

Abstract:Delayed neuronal death occured in the rat hippocampus following brief ischemia. We study and evaluate the effects of difluoromethyl ornithine (DFMO). ODC inhibitor, on the survival of neurons in the hippocampal CA_1 subfield. The pyramidal cell density in the CA_1 region is used as index of the effects. The results of experiment showed that DFMO has considerably reduced neuron loss in the CA_1 region. The favorabal effects are obviously dose-dependent and administration time-dependent. Furthermore, the results suggest that polyamine and ODC might play an importent role in the genesis of the delayed neuronal death following transient cerebral ischemia.
