摘    要:目的 探讨肝癌细胞多重耐药性形成的分子机制。方法 采用逐步培养法,诱导人肝癌细胞株HepG2产生耐药性,以MTT法测定其耐药性的程度,检测细胞表面MDR、MRP分子表达水平,并确定其与耐药性的关系。结果 耐药细胞表面MDR、MRP的表达明显增高,药物在细胞内的积聚减少;阻断MDR、MRP的功能,可明显逆转细胞的耐药性。结论 MDR、MRP的高表达是肝癌细胞形成多重耐药的性要机制之一。

Abstract:Objective] To investigate the roles of MDR, MRP in multidmg resistance of liver carcinoma cell lines. Methods] To induce the multidmg resistance, human hepatic carcinoma cell lines were cultured in medium containing gradually increased concentratioins of ADM. MTT, FCM were applied to measure the resisance of cell line to chemical agents, the concentrations of chemical agents inside the cells; MDR, MRP expressions on the cell membrane, respectively. Verapamil was given to restore the sensitivity of cell lines to chemical agents. Results] MDR,MRP markedly expressed on resistant cell lines HepG2/ADM, the intracellutar concentrations of chemical agentswere significantly reduced; blocking the MDR, MRP functions with verapamil could restore the sensitivity of cell lines to chemical agents. Conclusions] The high expressions of MDR, MRP were one of the important mechanisms of multidrug resistance of hepatic cell carcinoma.
