补中益气汤对A549/DDP肺癌荷瘤BALB/c小鼠耐药相关蛋白MRP表达的影响

目的研究补中益气汤对A549/DDP肺癌荷瘤BALB/c小鼠实体瘤中耐药相关蛋白(multidrug resistance-associated protein,MRP)表达的影响,探讨补中益气汤对A549/DDP的治疗作用机制。方法体外培养A549/DDP细胞,接种健康BALB/c小鼠,荷瘤成功后予补中益气汤治疗,实验分组为:荷瘤对照组,补中益气汤低剂量组及补中益气汤高剂量组,利用RT-PCR检测实体瘤MRP mRNA的表达,免疫组化法检测MRP蛋白表达。结果同荷瘤对照组相比,高、低剂量的补中益气汤均可降低耐药相关蛋白MRP的表达水平,且补中益气汤高剂量组效果同低剂量组相比无明显区别。结论补中益气汤的抗肿瘤作用可能与其降低MRP的表达相关。

Effect of Buzhongyiqi decoction on MRP expression in A549/DDP lung drug resistance tumor-bearing BALB/c mice

Objective To study the effect of Buzhongyiqi decoction on the multidrug resistance-associated protein（MRP ） expression in A549/DDP lung drug resistance tmnor-bearing BALB/c mice. Methods The A549/DDP hmg drug resistance tumor-bearing model was established by inoculation of A549/DDP cells into BALB/c mice subcutaneously. The mice were randomized into tumor-bearing mice control group, Buzhongyiqi decoction lower does group, Buzhongyiqi decoction higher does groups. Immunohistochemistry and RT-PCR methods were used to demonstrate the expression of MRP protein and MRP mRNA in A549/DDP tumors. Results The expression levels of MRP in A549/DDP tumor were decreased in both Buzhougyiqi decoction lower does group and higher does group than in tumor-bearing mice control group, with no difference between lower does group and higher does group. Conclusion The antitumor effect of Buzhongyiqi decoction might be related to the decrease of the expression level of MRP.
