槲皮素对去卵巢大鼠生殖系统及骨密度的影响

摘    要:目的探讨槲皮素在体内的雌激素样作用。方法健康雌性SD大鼠48只按体重随机分为6组:假手术组(SHAM)、单纯去卵巢组(OVX)、17β-雌二醇组ERT,0.1mg/(kg·d)]、高剂量槲皮素组QH,300mg/(kg·d)]、中剂量槲皮素组QM,150mg/(kg·d)]、低剂量槲皮素组QL,75mg/(kg·d)]。除假手术组外其余各组均切除双侧卵巢,术后1周开始给药,连续16周。给药期间每天观察阴道脱落细胞涂片。处死大鼠后,测量子宫重量计算子宫系数,病理切片观察子宫内膜及腺体,测量腰椎和股骨骨密度。结果槲皮素能使去卵巢大鼠阴道上皮细胞角化、子宫系数增加、腺体及内膜增生,骨密度增加(P〈0.05)。结论槲皮素在去卵巢大鼠体内具有弱雌激素样作用,能减轻雌激素缺乏导致的生殖系统萎缩,且能防止骨量丢失。

Abstract:Objective To study the estrogen-like effect of quercetin in vivo. Methods There were 48 adult female SD rats assigned to 6 groups:sham-operated group(SHAM) , ovariectomized group(OVX) , 1713-estradiol replacement therapy group ERT, 0. 1 mg/( kg·d ), sc] ; high-dose quercetin-treated group QH, 300 mg/( kg· d) ,ig] ; middle-dose quercetin-treated group QM ,150 mg/( kg·d) ,ig] ; low-dose quercetin-trea- ted group QL,75 mg/( kg·d) ,ig]. Except of SHAM group,rats in other groups were performed by bilateral ovariectomy. Administration started on 8th day after operation and lasted 16 weeks. During that time,we observed vaginal smear of rats everyday. Weighed uterus, observed endometria, and measured bone mineral density of femurs and lumbar vertebrae after executing rats. Results In ovarieetomized rats,quereetin hornified vaginal epithelial cells cornification,increased uterus coefficient and amount of glandular organs of uterus,thickened endometria,and prevented from decreasing of bone mineral density. Conclusion Quercetin had weakly estrogen-like effect,it could prevent reproductive system of rats from analosis because of hormonoprivia. Furthermore,it prevented from decreasing of bone mineral density in ovariectomized rats.
