摘    要:目的:建立乙型肝炎病毒(HBV)感染人胎盘滋养层细胞系Bewo的细胞模型,为HBV母婴传播过程中胎盘细胞感染机制的研究提供体外细胞模型。方法:选择人胎盘滋养层细胞系Bewo进行体外培养,用高浓度HBV DNA阳性血清(5×106/拷贝.ml-1)与Bewo细胞共培养48 h后,采用选择性PCR技术检测培养细胞中HBVrcDNA和HBVcccDNA。结果:HBV DNA阳性血清与Bewo共培养48 h,Bewo细胞中HBV rcDNA和HBV cccDNA均呈现阳性结果(243 bp和373 bp)。HBVDNA阳性血清与Bewo共培养对细胞形态无明显影响。结论:HBV能够直接感染人胎盘滋养层细胞系Bewo,该细胞系可用于进行HBV母婴传播过程中胎盘细胞感染机制的研究。

Abstract:Objective:To construct a model of human placental trophoblast Bewo cell lines infected by hepatitis B virus(HBV),provide an extracorporeal cell model for researching the mechanism of placental cell infection during the course of HBV maternal-neonatal transmission.Methods:Human placental trophoblast Bewo cell line was selected for culture in vitro,Bewo cells were cocultured with high dose positive HBV DNA sera(5×106/copy·ml-1) for 48 hours.Selective PCR technique was used to detect HBVrcDNA and HBVcccDNA in Bewo culture supernatant.Results:After 48 hours,positive HBVrcDNA and HBVcccDNA were found in Bewo cells(243 bp and 373 bp).Coculture of positive HBV DNA sera and Bewo cells had no obvious impact on cell morphology.Conclusion:HBV can infect human placental trophoblast Bewo cell line directly,which can be used to study the mechanism of placental cell infection during the course of HBV maternal-neonatal transmission.
