| 摘 要： | 目的探讨PTEN异常表达及其基因启动子区异常甲基化与甲状腺乳头状癌发生、发展及转移的关系。方法采用免疫组化、western-blot、甲基化特异性PCR(MSP)分析正常甲状腺与甲状腺乳头状癌组织中PTEN蛋白表达差异及PTEN基因启动子区甲基化情况。结果与正常甲状腺组织相比,甲状腺乳头状癌组织中PTEN蛋白表达明显下调(P<0.01),同时笔者发现PTEN基因蛋白的下调与肿瘤的淋巴结转移状况呈负相关(P<0.005),且甲状腺乳头状癌组织中PTEN基因启动子区甲基化程度显著增高。结论 PTEN蛋白低表达是甲状腺乳头状癌的发生、转移的重要原因之一,其低表达与基因启动子区过度甲基化有关。
