启膈散、沙参麦冬汤、通幽汤和补气运脾汤对人食管癌EC9706细胞增殖及凋亡的影响

目的观察启膈散、沙参麦冬汤、通幽汤、补气运脾汤各证方对人食管癌细胞株EC9706细胞增殖和凋亡的影响作用。方法体外培养人食管癌细胞株EC9706细胞,分别用启膈散、沙参麦冬汤、通幽汤和补气运脾汤处理细胞,倒置显微镜下观察EC9706细胞增殖和形态变化;MTT法检测各证方对EC9706细胞增殖影响的量效关系;TUENL法和荧光显微镜观察各证方诱导EC9706细胞凋亡的作用;Annexin V FITC/PI双标记流式细胞术检测各证方诱导EC9706细胞凋亡率：结果启膈散、沙参麦冬汤和通幽汤对EC9706细胞增殖的抑制率随药物浓度增加而上升,呈现剂量-效果依赖性关系,在药物浓度为6400μg/ml时,三组抑制率分别为78%、63%、78%。启膈散、沙参麦冬汤和通幽汤不同程度地诱导细胞发生凋亡,各组诱导细胞的凋亡率依次为：沙参麦冬汤组〉启膈散组〉通幽汤组。结论启膈散、沙参麦冬汤和通幽汤可不同程度地明显抑制人食管癌细胞株EC9706细胞增殖,补气运脾汤对体外培养细胞增殖的直接抑制作用很弱。启膈散、沙参麦冬汤和通幽汤均可不同程度诱导人食管癌EC9706细胞凋亡。

Effects of Qige Power,Shashenmaidong Decoction,Tongyou Decoction and Buqiyunpi Decoction on the Proliferation and Apoptosis of Human Esophageal Carcinoma Cell Line EC9706

Objective Effects of Qige Power,Shashenmaidong Decoction,Tongyou Decoction and Buqiyunpi Decoction on the cell proliferation and apoptosis are studied to explore the mechanism for treating esophageal cancer.Methods After treating human esophageal carcinoma cell line EC9706 with Qige Power,Shashenmaidong Decoction,Tongyou Decoction and Buqiyunpi Decoction,growth and morphological change are observed under inverted microscope,cell proliferation is assayed by MTT.Apotosis is observed by TUENL method and under fluoromicroscope,cell apoptosis ratio is measured by FCM using fluorescent dye.Results Qige Power,Shashenmaidong Decoction,Tongyou Decoction could inhibit cell growth in different degree,the percentage of inhibition are separately79%,63%and 78%while the medicine consistence is 6400μg/ml.Qige Power,Shashenmaidong Decoction,Tongyou Decoction could induce the cell apoptosis differendy in which effect of Shashenmaidong Decoction was the best,the second was Qige Power and the last was Tongyou Decoction.Conclusion Qige Power,Shashenmaidong Decoction,Tongyou Decoction inhibit cell growth in different degree in different degree,the inhibition of Buqiyunpi Decoction on cultured carcinoma cell growth is very weak.Qige Power,Shashenmaidong Decoction,Tongyou Decoction inhibit cell growth in different degree by inducing the cell apoptosis
