卢帕他定对大鼠油酸型急性肺损伤的保护作用研究

摘    要: 目的探讨卢帕他定(rupatadine)对大鼠油酸型急性肺损伤(acute lung injury,ALI)的作用特性及其对肿瘤坏死因子-α(TNF-α)表达的影响。方法制备油酸型大鼠急性肺损伤模型,利用动物肺功能测定仪测定肺损伤过程中吸气气道阻力(IR)、呼气气道阻力(ER)和动态肺顺应性(Cdyn)的变化并分析诸项指标变化的意义、称重法计算湿/干重比(W/D),紫外分光光度法检测肺泡灌洗液(BALF)中蛋白含量,酶联免疫吸附法(ELISA)检测TNF-α含量。结果卢帕他定可显著改善油酸型肺损伤大鼠的肺功能指标,降低肺组织湿/干重比,减少肺泡灌冼液中蛋白含量,抑制TNF-α的升高,抑制率达到24.22%。结论卢帕他定时大鼠油酸型肺损伤有明显的保护作用且具剂量依赖性,其作用机制可能与降低ALI时肺组织中TNF-α炎症因子合成与释放有关

Abstract:OBJECTIVE To investigate the effect of rupatadine on acute lung injury induced by oleic acid in rats.METHODS Rats were randomly divided into six groups.The changes of lung inspiratory resistance,expiratory resistance and dynamic lung compliance were recorded.BALF(bronchoalveolar lavage fluid)protein was measured by spectrophotometer and TNF-αlevel was examined by En- zyme linked immunosorbent assay(ELISA).Lung wet tu dry weight ratio was calculated to assess alveolar edema.RESULTS The pre- treatment of rupatadine significantly alleviated oleic acid-induced lung injury accompanied by the imprnvement of lung function aud reduc- tion of lung wet tn dry weight ratio.Furthermore,rupatadine also decrcased the concentration of protein in BALF and inhibited the in- crease of the TNF-αsignificantly.CONCLUSION Rupatadine has a dose-depeudent beneficial effect on acute lung injury induced by oleic acid in rats.The ultimate reduction of inflammatory factor was involved,at least in part,in the mechanism of rupatadine effects.
