摘    要:目的:探讨益肾活血胶囊提取液(YSHXC)对高同型半胱氨酸(HCY)致兔血管平滑肌细胞(VSMCs)核因子-κB(NF—κB)活化及血管细胞黏附分子-1(VCAM—1)表达的影响。方法:以HCY诱导体外培养的兔主动脉平滑肌细胞建立细胞增殖模型。实验分为正常组、HCY组、HCY+YSHXC(小、中、大)剂量组。通过细胞计数观察VSMCs的增殖情况,同时采用免疫组化和RT—PCR技术分别检测NF—κB、vCAM—1的蛋白及mRNA表达。结果:HCY诱导后可使NF—κB活化、VCAM—1的表达增强;而YSHXC可从蛋白和mRNA水平明显抑制其表达(P〈0.01),并呈现剂量依赖关系,这在各实验组细胞数目也有相应的体现。结论:YSHXC能明显抑制HCY诱导的NF—κB活化及VCAM—1的表达,减少VSMCs的增殖和向内皮的迁移,可能对延缓动脉粥样硬化(AS)的发生发展起到一定作用。

Abstract:Objective: To investigate the effects of "Yishen Huoxue Capsule"(YSHXC)on NF-κB and VCAM-1 of rabbits' VSMCs induced by HCY. Methods: Cultured cells proliferation model of rabbits' VSMCs were established with HCY in vitro. Five groups were set up as follows: blank group, model group, low-dose YSHXC group, middle-dose YSHXC group and high-dose YSHXC group. The VSMCs proliferation was determined by cell counting. The expressions of protein and mRNA of NF-κB and VCAM-1 were detected with immunohistocbemistry and RT-PCR respectively. Results: HCY induced the activating of NF-κB as well as the expression of VCAM-1. YSHXC significantly reduced the expression of protein and mRNA of NF-κB and VCAM-1 in a dose-dependent manner, which can also be reflected from the cells counting. Conclusion: YSHXC can significantly inhibit the activation of NF-κB and the expression of VCAM-1 induced by HCY, reduce the VSMCs' proliferation and its migration to the endothelium, thus may delay the occurrence and development of artberosclerosis(AS).
