胃癌细胞AGS中KDR的表达对人脐静脉内皮细胞生物学行为的影响

摘    要:目的观察胃癌细胞AGS中血管内皮生长因子受体KDR表达水平发生变化时,其培养上清对人脐静脉内皮细胞(HUVECs)生物学行为的影响。方法采用Western blot及免疫细胞化学的方法检测AGS细胞中KDR的表达;利用重组腺病毒介导的KDR siRNA转染AGS细胞,通过细胞增殖试验、迁移试验及管状结构形成试验,观察不同条件培养基作用下内皮细胞生物学特性的变化。结果胃癌细胞AGS表达KDR,转染siRNA后表达水平降低;条件培养基影响了HUVECs增殖、迁移、形成管状结构的能力,转染siRNA的AGS细胞或亲本细胞组较RPM11640组均增强,前者更强。结论胃癌细胞AGS来源的条件培养基对内皮细胞的增殖、迁移、管状结构的形成具有促进作用,抑制AGS细胞中KDR的表达,这种作用被增强。

Abstract:Objective To investigate the effects of c on ditioned mediums derived from gastric cancer cells AGS with different KDR levels on the biological behavior of HUVECs. Methods The expres sion of KDR in AGS cells was detected by western blot and immunological cytochem istry. The recombined adenoviral-mediated siRNA targeted KDR was used to transfe ct AGS cells, and the effects of different conditioned mediums on the properties of HUVECs were observed with proliferation assay, migration assay and tube form ation assay. Results AGS cells expressed KDR, and the exp ression was reduced after the transfection of siRNA. The proliferation, migratio n and tube formation abilities were promoted by the mediums derived from siRNA t ransfected-AGS cells or parent-AGS cells compared with RPMI1640, and the former was stronger. Conclusion The conditioned medium derived from AGS cells promoted the proliferation, migration and tube formation abilities of HUVECs. Reducing the expression of KDR in AGS cells, this action became str onger.
