孕中期羊水干细胞的生物学特性的研究

摘    要:目的对孕中期羊水标本进行体外分离、培养、扩增,研究孕中期羊水来源干细胞(AFC)的生长特性、细胞表面标记。方法采用贴壁法体外分离孕中期羊水来源干细胞,绘制细胞生长曲线;流式细胞仪检测细胞表面标记。结果原代培养细胞生长潜伏期4-6d,对数增殖期7-13d,生长平台期14-18d;传代培养细胞生长潜伏期1-2d,对数增长期3-6d,生长平台期7-10d。AFCs均一表达CD44,不表达CD45。结论从人孕中期羊水中成功分离具有干细胞性质的细胞群,在体外可迅速扩增。

Research on bioiogicfal features of human amniotic fluid-derived stem cells in second-trimester

Abstract:Objective Human amniotic fluid samples in second trimester. To study the growth pattern and surface markers of human amniotic fluid derived stem cells in second-trimester. Methods The growth curves of the AFCs were drawn. The flow cytometry was used to detect surface antigenic markers. Results The latent phase of the primary culture cells was 4 6days,followed by 7 13 clays of logarithmical proliferation; the plateau began at day 14 18. The sub cultures showed 1-2 days of latent phase, followed by 3-6 days of logarithmical proliferation, then entered the plateau (days) 7 10. The AFCs expressed positive CD44 unanimously,but don't expressed CD45. Conclusion Experiment has successfully isolated stem cells from second trimester amniotie fluid. These stem cells can he expanded in vitro rapidly.
