Effect of Yangxin Decoction on Vascular Endothelial Dysfunction Induced by Homocysteine in Rats

Abstract:Objective:To establish vascular endothelial dysfunction model in SD rats fed with high methionine diet,to observe the effect of Yangxin Decoction on vascular endothelial dysfunction and explore its related mechanism.Methods:40 male SD rats were randomly divided into blank control group,model group,folic acid group and Yangxin Decoction group,with 10 rats in each group.The blank control group was fed with normal diet,and the model group,folic acid group and Yangxin Decoction group were fed with normal diet containing 3%L-methionine for 12 weeks.After successful establishment of the model,rats in the blank control group and the model group were given the same volume of purified water by gavage;rats in the folic acid group were given folic acid tablet suspension 1.8 mg/(kg·d),and rats in the Yangxin Decoction group were given Yangxin Decoction extract suspension 2.34 g/(kg·d)by gavage for 4 weeks.At the end of the experiment,the rats were anesthetized and weighed,and the thoracic aorta was taken to observe the ultrastructural changes of vascular endothelium under electron microscope;the blood of abdominal aorta was centrifuged,and the serum levels of no,ET-1,Hcy,sEPCR and STM were detected by ELISA.Results:Under the electron microscope,the vascular endothelium of the blank control group was intact without obvious damage.Compared with the blank control group,most of the vascular endothelium of the model group fell off,occasionally residual endothelium was found,and the degree of damage was significantly aggravated.Compared with the model group,the vascular endothelium of the Yangxin Decoction group was basically intact,and the degree of damage was significantly reduced Compared with the control group,the level of serum NO in the model group decreased,and the levels of ET-1,Hcy,s EPCR and STM increased(P<0.01);compared with the model group,the level of serum NO in the Yangxin Decoction group increased,and the levels of ET-1,Hcy,s EPCR and STM decreased(P<0.01).Conclusion:Yangxin Decoction can increase serum NO level,decrease serum ET-1,Hcy,sEPCR and STM levels,and improve vascular endothelial dysfunction and pathological injury.
