血红素加氧酶在局灶性脑缺血再灌注大鼠大脑组织表达测定

摘    要:目的]测定血红素加氧酶(HO)在局灶性脑缺血再灌注大鼠大脑组织表达。方法]使用随机平行对照方法,将SPF级雄性大鼠20只随机分为两组,缺血组15只参照Zea-logue方法造大鼠大脑中动脉缺血2h再灌注24h模型,假手术对照组5只采用同样方法暴露颈动脉后缝合切口。观察脑组织形态结构,测定HO-1表达。结果]假手术组脑组织形态结构正常,模型组大鼠大脑缺血区组织明显水肿。HO-1表达相同部位缺血组明显高于假手术组(P<0.01)。结论]局灶性脑缺血再灌注大鼠大脑血HO升高,HO-1参与了局灶性脑缺血再灌注损伤。

A Study on Expression and Significance of HO-1 in Cerebraltissues of Rat after Focal Cerebral Ischemia and Reperfusion

Abstract:Objective] To investigate the effects of the expression of HO-1 in cerebral tissues of rat after focal cerebral ischemia and reperfusion.Methods] 2 hours right intraluminal middle cerebral artery occlusion with reperfusion was induced in 20 rats,and the specimen of brain were performed at the point after 24 hours reperfusion;expressions of HO-1were detected by genechip profile and RT-PCR method.Results] Expressions of HO-1 were seen sparsely in the control specimens,and were markly increased afer transient focal cerebal ischemia and reperfusion(P<0.01).Conclusion] Expression of HO-1 is a significant response to transient focal cerebral ischemia/reperfusion and contributes to the early formasion of vasogenic edema that can be observed all parts of brains.
