肺鳞癌与人乳头瘤病毒及p53表达的关系

摘    要:目的 探讨肺鳞癌组织中人乳头瘤病毒(HPV)感染与p53蛋白表达的关系并分析作用机制。方法 收集84例肺鳞癌和18例支气管粘膜慢性炎症标本,采用地高辛标记的HPV-DNA探针,用原位杂交法检测组织中的HPV-DNA存在情况,并用免疫组化SABC法检测肺鳞癌组织中p53及HPVE6,E7蛋白的表达,将二者进行对照观察。结果 ①HPV原位杂交阳性信号位于细胞核内,肺鳞癌HPV检出率为49%,对照组慢性炎为6%,差异显著。②肺鳞癌组织中存在HPVE6,E7蛋白表达,免疫组化方法与原位杂交方法结果相符。③84例肺鳞癌中30例检出p53蛋白表达,其中17例HPV阳性,13例HPV阴性,HPV阳性组与阴性组差异不显著。结论 肺鳞癌与HPV感染关系密切,肺鳞癌组织中也存在p53基因表达,p53蛋白积聚与HPV感染无明显相关性。

Abstract:AIM To investigate the relationship between human papillomavirus (HPV) infection and the accumulation of p53 in squamous cell carcinoma (SQCC) of lungs. METHODS 84 of SQCC and 18 of bronchial chronic inflam mation samples were detected by in situ hybridization for human papillomavirus using digoxingemin labelled DNA probe. HPV E6, E7 protein and the accumulation of p53 were revealed by SABC immunohistochemistry. RESULTS The positive signal was mainly located in the nuclei of the cells. 49% of SQCC and 6% of chronic inflammation tissues were positive for HPV DNA by in situ hybridization; The result of in situ hybridization was confirmed by the detection of E6 and E7 protein with SABC immunohistochemistry. 30 of the 84 squamous cell carcinomas were positive for p53 and no significant difference between HPV positive and HPV negative groups was found. CONCLUSION HPV DNA was closely associated with SQCC, and there was accumulation of p53 in SQCC, but the relationship between HPV DNA and accumulation of p53 was not found.
