大孔树脂纯化马兰总三萜工艺研究

摘    要:目的 研究大孔树脂纯化马兰总三萜的工艺。方法 以齐墩果酸为对照品,以吸附量和解吸率作为效果评定指标,筛选出对马兰中总三萜纯化效果最好的大孔树脂,并对上样量、上样液浓度、洗脱速率、洗脱乙醇浓度及用量参数进行考察,筛选出最佳工艺条件。结果 HPD-500的吸附量为0.495 5 mg/mL,解吸率为30.47%,效果最佳;最佳上样液浓度:0.02 g/mL;流速:2 mL/min;洗脱剂:70%乙醇;洗脱体积:2倍柱体积。纯化后马兰三萜含量可达到17.81%。结论 HPD-500大孔树脂在所得工艺条件下可以对马兰中总三萜进行较好的纯化。

Purification Process of Total Triterpenes from Kalimeris indica Using Macroporous Resin

Abstract:Objective To investigate the purification process of total triterpenes from Kalimeris indica using macroporous resin. Methods With oleanolic acid as the reference substance and adsorption quantity and desorption rate as evaluation indices, the macroporous resin with the best purification effect on total triterpenes was screened out. The optimal process conditions were screened out by investigating the sample loading quantity, sample liquid concentration, elution rate, and alcohol concentration and volume for elution. Results HPD-500 had an adsorption quantity of 0.495 5 mg/mL and a desorption rate of 30.47% and had the best effect. The optimal process conditions were a sample liquid concentration of 0.02 g/mL, a flow rate of 2 mL/min, an eluent of 70% alcohol, and an elution volume of two-fold column volume. The content of triterpenes after purification reached 17.81%. Conclusion Good purification of total triterpenes in Kalimeris indica Sch. can be achieved by HPD-500 macroporous resin under the above conditions.
