脑缺血再灌注大鼠海马区神经营养因子的表达及桃红四物汤的干预

摘    要:目的 观察脑缺血再灌注后大鼠海马区不同时间神经营养因子表达的变化及桃红四物汤的干预作用。方法 采用大脑中动脉线栓法构建大鼠局灶性脑缺血再灌注模型,将实验动物随机分为假手术组、模型组、桃红四物汤组和尼莫地平组。采用TTC染色法检测脑梗死体积,酶联免疫吸附法测定大鼠海马区脑源性神经营养因子(brain derived neurotrophic factor, BDNF)、神经生长因子(nerve growth factor,NGF)、神经营养因子3(neurotrophin-3,NT-3)的表达水平。结果 与假手术组相比,模型组术后12 h、24 h、7 d、14 d脑梗死体积明显增大。与模型组相比,桃红四物汤组在术后7 d和14 d脑梗死体积显著减小,尼莫地平组术后12 h、24 h、7 d和14 d脑梗死体积显著减小。与假手术组相比,模型组术后12 h海马区BNDF表达水平显著提高,术后24 h海马区NGF表达水平显著提高,各时点NT-3表达水平的差异无统计学意义;与模型组比较,术后7 d和14 d,桃红四物汤组、尼莫地平组BDNF表达水平显著提高,术后24 h和7 d,尼莫地平组NGF表达水平显著提高,桃红四物汤组NGF和NT-3各个时间点的表达水平无明显差异。结论 大鼠脑缺血再灌注一定时间后海马区BDNF和NGF的表达水平增高,有利于受损的神经细胞恢复,桃红四物汤通过促进BDNF的表达而保护神经细胞。

Expression of Neurotrophic Factor in the Hippocampus of Rats with Cerebral Ischemia/Reperfusion and Interventional Effect of Taohong Siwu Decoction

Abstract:Objective To investigate the change in the expression of neurotrophic factor in the hippocampus of rats at different time points after cerebral ischemia/reperfusion and the interventional effect of Taohong Siwu Decoction. Methods Middle cerebral artery occlusion was performed to establish a rat model of focal cerebral ischemia/reperfusion. The experimental animals were divided into sham-operation group, model group, Taohong Siwu Decoction group, and nimodipine group. TTC staining was used to observe cerebral infarct volume, and ELISA was used to measure the expression of brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and neurotrophin-3 (NT-3) in the hippocampus of rats. Results Compared with the sham-operation group, the model group had a significant increase in cerebral infarct volume at 12 hours, 24 hours, 7 days, and 14 days after surgery. Compared with the model group, the Taohong Siwu Decoction group had a significant reduction in cerebral infarct volume at 7 and 14 days after surgery, and the nimodipine group had a significant reduction in cerebral infarct volume at 12 hours, 24 hours, 7 days, and 14 days after surgery. Compared with the sham-operation group, the model group had significant increases in the expression of BDNF in the hippocampus at 12 hours after surgery and the expression of NGF in the hippocampus at 24 hours after surgery and had no significant changes in the expression of NT-3 at each time point. Compared with the model group, the nimodipine group and the Taohong Siwu Decoction group had a significant increase in the expression of BDNF at 7 and 14 days after surgery; the nimodipine group had a significant increase in the expression of NGF at 24 hours and 7 days after surgery; the Taohong Siwu Decoction group had no significant changes in the expression of NGF and NT-3 at each time point. Conclusion The expression of BDNF and NGF in the hippocampus of rats increases at a certain time point after cerebral ischemia, which helps with the recovery of damaged neurons. Taohong Siwu Decoction protects neurons by promoting the expression of BDNF.
