咪喹莫特对人表皮朗格汉斯细胞及HaCaT细胞分泌细胞因子的影响

摘    要:目的 研究咪喹莫特对人表皮朗格汉斯细胞(LC)及HaCaT细胞细胞因子分泌水平的影响。方法 联用密度梯度离心和免疫磁珠的方法分离纯化LC,以5μg/mL咪喹莫特刺激LC及HaCaT细胞,4h后取培养上清,用ELISA方法检测肿瘤坏死因子α、白介素1β及白介素6的含量。结果 LC咪喹莫特处理的实验组培养上清中肿瘤坏死因子α、白介素1β及白介素6含量均较未处理对照组明显增高(P均<0.05)。HaCaT细胞3种细胞因子分泌量在实验组与对照组中差异无统计学意义(P均>0.05)。结论 咪喹莫特能增加LC细胞因子肿瘤坏死因子α、白介素1β及白介素6的分泌,但对HaCaT细胞上述3种细胞因子的分泌未见明显影响。

Abstract:Objective To investigate the effects of imiquimod on cytokine secretions of normal human epidermal Langerhans cells (LCs) and HaCaT cells, and to better understand the mechanism of immune regulation by imiquimod. Methods LCs were sorted from prepared epidenmal cells by density gradient centrifugation and magnetic-activated cell sorting (MACS). LCs and HaCaT cells were cultured in media with or without 5 μg/mL of imiquimod for 4 hours, then cell-free culture supernatants were harvested, cytokines were detected by ELISA kits. Results TNF-α, IL-1β and IL-6 secreted from imiquimod treated LCs were all higher than those from control LCs (all P<0.05), but there is no statistical difference between the cytokines from imiquimod treated HaCaT cells and those from control HaCaT cells. Conclusion Imiquimod can upregulate the secretions of cytokines from human epidermal LCs but not from HaCaT cells.
