米非司酮与皮质醇合用对大鼠睾丸间质细胞分泌睾酮的影响

摘    要:为研究米非司酮和皮质醇对大鼠睾丸间质细胞(Leydig Cells)的睾酮基础分泌及促性腺激素刺激睾酮分泌的影响,将雄性大鼠处死,取睾丸间质细胞进行体外培养,用放射免疫分析方法测定睾酮含量。实验结果显示米非司酮抑制睾酮的基础分泌,而皮质醇对其无影响;两药单独使用时,均抑制促性腺激素刺激的睾酮分泌,合用有协同作用。本研究表明米非司酮和皮质醇均能影响睾丸的甾体激素的合成。

Effect of Combination of Mifepristone with Cortisol on the Testosterone Production of Rat Leydig Cells in Vitro.
Abstract:Objectives:To investigate the effect of mifepristone and cortisol on basal or hCG-stimulated testosterone production. Methods : Adult Wistar rats were sacrificed and Leydig cells collected for incubation with mifepristone, cortisol and hCG. T levels in culfurals was measured by radioim-munoassay. Results: Incubation with mifepristone decreased basal T production in a dose-dependent manner,but cortisol didn't affected it,Both mifepristone and cortisol inhibited hCG-stimulated T production and they have synergism. Conclusions : These data suggested that cortisol play an important role in the testicular steroidgenesis. mifepristone and cortisol were found to have a direct effect on testosterone production of Leydig cells in rats.
