川芎水提取物与川芎嗪对氯化钴诱导的缺氧内皮细胞的作用比较

摘    要:目的:研究川芎水提取物与川芎嗪对缺氧人脐静脉内皮细胞(EAHY926)的影响,比较二者对内皮细胞的保护作用。方法:体外培养EAHY926,采用化学诱导缺氧建立模型,用川芎水提取液及川芎嗪干预后,测定细胞活力,并检测丙二醛(malonic acid,MDA)含量、超氧化物歧化酶(superoxide dismutase,SOD)及乳酸脱氢酶(lactate dehydrogenase,LDH)的变化,比较二者的作用效果。结果:采用单因素方差分析及SNK检验,与模型组相比,川芎水提取物组及川芎嗪参组均能显著提高细胞活力(P0.05);与正常对照组比较,模型组MDA显著升高(P0.01),且川芎水提取液组、川芎嗪组与正常组比较无明显差异;与模型组比较,川芎水提物组及川芎嗪组LDH外漏量显著降低(P0.01),川芎水提物组细胞及川芎嗪组MDA含量有显著差异(P0.01),川芎水提物组细胞SOD活性显著升高(P0.05);川芎水提取物组与川芎嗪组比较,LDH外漏量有明显差异(P0.01)。结论:川芎水提取物及川芎嗪能显著拮抗缺氧对EAHY926的损伤,且川芎水提取物优于川芎嗪。

Abstract:Objective:To study the influence of Chuanxiong(Rhizoma Chuanxiong)water extract and ligustrazine on EAHY926 respectively,and compare their functions in protecting endothelial cells.Methods:EAHY926 was cultured in vitro,the hypoxia model was established by using chemical for induction.After being interfered by Chuanxiong(Rhizoma Chuanxiong)water extract and ligustrazine respectively,cell viability was determined,malondialdehyde (MDA) content,and lactate dehydrogenase (LDH) changes were tested.The effects of the two groups were compared.Results:By using one way ANOVA and SNK test,compared with the model group,both Cbuanxiong water extract and ligustrazine groups could significantly improve the cell vitality (P < 0.05).Compared with the nornal control group,the model group's MDA increased significantly (P < 0.01).There were no significant differences between Chuanxiong water extract group and ligustrazine group and the normal group.Compared with the model group,Chuanxiong water extract and ligustrazine groups' LDH leakage were significantly reduced (P < 0.01).Chuanxiong water extract group cell and ligustrazine group MDA content had significant difference (P < 0.01).Chuanxiong water extract group activity of SOD increased significantly (P < 0.05);LDH leakage amount had significant difference between Chuanxiong water extract and ligustrazine groups (P < 0.01).Conclusion:Chuanxiong water extract and ligustrazine can significantly resist damage on EAHY926 due to hypoxia,and Chuanxiong water extract is better than ligustrazine.
