生脉饮对慢性心力衰竭大鼠心功能及血清游离脂肪酸的影响

摘    要:目的:探讨生脉饮对慢性心力衰竭(chronic heart failure,CHF)大鼠心功能及血清游离脂肪酸的影响。方法:采用结扎大鼠冠状动脉前降支法造成心肌梗死后形成CHF模型。于手术4周后用GEL-400彩色超声心动仪进行心功能检测,以左室射血分数(left ventricular ejection fraction,LVEF)下降到60%以下作为心力衰竭模型成功标志。将造模成功的大鼠随机分为模型组、卡托普利组、生脉饮高剂量组、生脉饮低剂量组及正常对照组,每组各10只。对照组不进行药物干预,常规饲养。模型组每日灌胃与治疗组相同体积的生理盐水,卡托普利组取卡托普利按50 mg·(kg·d)-1灌胃给药,生脉饮高剂量组取生脉饮颗粒剂按1.6 g·(kg·d)-1灌胃给药,生脉饮低剂量组取生脉饮颗粒剂按0.8g·(kg·d)-1灌胃给药。超声心动测定大鼠心功能,包括测量左室舒张末期内径(left ventricular end diastolic diameter,LVDd),左室收缩末期内径(left ventricular end systolic diameter,LVDs),LVEF,左室短轴缩短率(left ventricular fractional shortening,FS)。采用酶比色法检测血清中FFA浓度。结果:给药后模型组、生脉饮低剂量组LVDd、LVDs较给药前增加,差异具有统计学意义(P0.05),卡托普利组、生脉饮高剂量组给药后LVDd、LVDs均较给药前减少,但差异无统计学意义(P0.05)。模型组、生脉饮低剂量组给药后FS较给药前下降,差异具有统计学意义(P0.05),卡托普利组、生脉饮高剂量组给药后FS较给药前有所增加,但差异无统计学意义(P0.05);给药后各组与对照组比较,差异具有统计学意义(P0.05)。模型组给药后EF较给药前降低,余治疗组给药后均较给药前提高,但差异无统计学意义(P0.05),给药后生脉饮高剂量组与对照组比较,差异无统计学意义(P0.05),其余各治疗组给药后与对照组比较,差异有统计学意义(P0.05)。与对照组比较,各组大鼠血清FFA含量明显降低,差异具有统计学意义(P0.05),但各组间比较,差异无统计学意义(P0.05)。结论:生脉饮具有改善心力衰竭大鼠心脏收缩功能、增加射血能力的作用,从而增强心功能治疗心力衰竭,以生脉饮高剂量组效果更为明显。生脉饮高剂量组对心力衰竭大鼠血清FFA含量有一定增加的趋势。生脉饮可能通过干预大鼠心肌能量代谢能量底物的利用而发挥改善心功能的作用。

The Influence of Pulse-Engendering Decoction on Heart Function and Serum Free Fatty Acid of Rats with Chronic Heart Failure

Abstract:Objective:To probe into the influence of Pulse-Engendering Decoction on heart function and serum free fatty acid of rats with chronic heart failure (CHF).Methods:CHF model was established by ligation of anterior descending branch of coronary artery in rats.After 4 weeks of operation,cardiac function was detected by GEL-400 color Doppler echocardiography,and the left ventricular ejection fraction(LVEF) decreased to below 60% was regarded as a sign of heart failure model.Then those rats were randomly divided into the model group,Kato Pury group,Pulse-Engendering Decoction high-dose group,Pulse-Engendering Decoction low-dose group and the normal control group,with 10 rats in each group.The control group did not take drug intervention,but adopted conventional feeding.The model group was administered daily with the same volume of normal saline as the treatment group.Kato Pury group was given medicine for garage according to 50 mg · (kg · d)-1.Pulse-Engendering Decoction high-dose group was given medicine for gavage according to1.6 g · (kg · d)-1,while Pulse-Engendering Decoction low-dose group was given medicine for gavage according to 0.8 g · (kg · d)-1.Cardiac function was measured in rats by echocardiography,including left ventricular end diastolic diameter (LVDd),left ventricular end systolic diameter (LVDs),left ventricular ejection fraction (LVEF),and left ventricular fractional shortening(LVFS).The FFA concentration in serum was detected by enzyme colorimetric assay.Results:After taking medicine,LVDd and LVDs of the model group and Pulse-Engendering Decoction low-dose group increased compared with before administration,the difference was statistically significant (P < 0.05),while those of Kato Pury group and Pulse-Engendering Decoction low-dose group decreased compared with before administration,but the difference was not statistically significant(P > 0.05).After taking medicine,FS of the model group and Pulse-Engendering Decoction low-dose group decreased compared with before administration,the difference was statistically significant (P < 0.05),while those of Kato Pury group and Pulse-Engendering Decoction low-dose group increased compared with before administration,but the difference was not statistically significant (P > 0.05).Compared with the control group,the difference between the two groups was statistically significant (P < 0.05).After taking medicine,EF of the model group decreased,but EF of the other groups increased,but the difference was not statistically significant (P > 0.05);The difference between Pulse-Engendering Decoction high-dose group and the control group was not statistically significant(P >0.05);But the difference between each of the other groups and the control group was statistically significant(P > 0.05).Compared with the control group,the content of serum FFA in each group was significantly lower,the difference was statistically significant(P < 0.05),but there was no significant difference between the two groups (P > 0.05).Conclusion:Pulse-Engendering Decoction can improve heart failure rat cardiac systolic function,increase ejection ability,so as to improve cardiac function.Pulse-Engendering Decoction high-dose group has better efficacy,which can increase the content of FFA in serum of rats with heart failure.Pulse-Engendering Decoction can improve heart function by using the intervention of myocardial energy substrate metabolism in rats.
