糖痹康对糖尿病周围神经病变大鼠氧化应激的影响

摘    要:目的:观察中药糖痹康对DPN大鼠血清SOD、NO和NGF的影响,从而探讨其对DPN大鼠的抗氧化效果。方法:健康SD大鼠80只随机选取10只为正常对照组,普通饲料喂养,其余高脂饲料喂养,8周后STZ造模:用0.1 mmol·L~(-1)无菌枸橼酸钠缓冲液(PH 4.5,4℃),将STZ配成0.45%的溶液,按45 mg·kg~(-1)的剂量标准一次性给大鼠左下腹腔内注射。72 h后用血糖仪测尾尖血血糖,选持续血糖水平≥16.7 mmol·L~(-1)以上且血糖稳定3 d者为造模成功,按体质量随机进入各治疗组,模型复制成功后,按体质量随机分为以下5组,每组10只:模型组、弥可保组(西药组)、糖痹康低剂量组、糖痹康中剂量组、糖痹康高剂量组,加上正常组,共6组。糖痹康低剂量组:按成人剂量5倍给药,即生药4.175 mg·(kg·d)~(-1);糖痹康中剂量组:按成人剂量10倍给药,即生药8.35 mg·(kg·d)~(-1);糖痹康高剂量组:按成人剂量20倍给药,即生药16.7 mg·(kg·d)~(-1),均换算成糖痹康为3 m L灌胃;弥可保组:按成人剂量10倍给药,即1.97 mg·(kg·d)~(-1),用蒸馏水配成混悬液3 m L灌胃;模型组及正常组给予等量蒸馏水灌胃。采用ELISA法测定实验大鼠血清SOD、NO、NGF酶活性,按SOD、NO、NGF ELISA试剂盒说明操作。结果:与模型组相比较,糖痹康各剂量组有显著升高,高剂量组效果优于对照组;糖痹康各剂量组可以明显提高血清NGF水平。结论:中药糖痹康可以升高血清SOD、NO和NGF水平,抑制体内过高的氧化应激水平,阻止和/或延缓了DPN的发生和发展。

Abstract:Objective:To observe the influenee of Tangbikang on the levels of SOD,NO and NGF in rats with diabetic peripheral neuropathy (DPN) and explore the antioxidant effect on DPN rats.Methods:Ten rats among the 80 healthy SD rats were selected as normal control group fed with normal diet and the rest of the rats were fed with high-fat diet.After 8 weeks,STZ models were established.STZ were made into 0.45% solution by sterile sodium citrate buffer solution (pH 4.5,4℃).Rats were given left intraperitoneal injection with standard dosage of45mg/kg.After 72h,the tail-tip blood glucose was detected with glucose meter.If the continuous blood glucose level ≥ 16.7mmol/L and kept for 3d,the rats were successful established models.The rats were randomized into 5 groups according to the body mass,10 cases in each group:Model group,Mecobalamin group (western medicine group),Tangbikang low dosage group,Tangbikang medium dosage group and Tangbikang high dosage group.Tangbikang low dose group were given administration of 4.175mg/kg/d (5 times of adult dosage);Tangbikang medium dosage group were given administration of 8.35mg/kg/d (10 times of adult dosage);Tangbikang high dosage group were given administration of 16.7 mg/(kg · d)-1 (20 times of adult dosage);these three groups were intragastric administration replaced by Tangbikang of 3 mL;Meeobalamin group were given administration of 1.97 mg/(kg · d)-1(10 times of adult dosage) and intragastrie administration of suspension made by distilled water (3 mL);model group and normal group were given intragastric administration of distilled water of equal quantity.The enzyme activities of SOD,NO and NGF in the serum of rats were detected by ELISA.Results:Compared with model group,Tangbikang groups of low,medium and high dosage improved significantly;the effect in Tangbikang group of high dosage was superior to that in control group.Tangbikang groups of low,medium and high dosage can obviously increase the level of NGF.Conclusion:The Chinese medicine Tangbikang can improve the levels of SOD,NO and NGF,inhibit the excessive oxidative stress level,prevent and delay the occurrence and development of DPN.
