茵陈、大黄联合用药对四氯化碳肝损伤小鼠的保护作用

摘    要:目的:比较不同配比茵陈、大黄(茵黄)联合用药对四氯化碳急性肝损伤小鼠的保护作用,探究茵黄联合用药治疗小鼠四氯化碳急性肝损伤的最佳比例。方法:将80只小鼠随机分为空白组、模型组、阳性对照组、茵黄10∶0组、茵黄9∶1组、茵黄3∶1组、茵黄3∶2组、茵黄1∶1组、茵黄1∶9组、茵黄0∶10组,每组各8只。阳性对照组以联苯双酯(0.2 g·kg~(-1))进行灌胃,治疗组按上述比例灌胃给药;空白组、模型组分别以等量蒸馏水进行灌胃。各给药组连续灌胃给药7 d,末次给药1 h后,给药组与模型组均腹腔单次注射质量分数0.1%四氯化碳花生油溶液0.01 mL·g~(-1)。16 h后摘眼球取血,检测血清中谷丙氨酸氨基转移酶(alanine transaminase,ALT)、天冬氨酸氨基转移酶(aspartateaminotransferase,AST)水平。解剖取肝脏称重,计算肝体指数;制成肝组织匀浆,测定三酰甘油(three acyl glycerin,TG)、血清总胆固醇(total cholesterol,TC)浓度。取相同部位肝组织制片,HE染色后观察其病理学形态。结果:与模型组比较,茵黄不同比例治疗组均能降低肝损伤小鼠的肝体指数,差异具有统计学意义(P0.05)。与空白组比较,模型组ALT、AST水平显著上升;与模型组比较,茵黄10∶0组、茵黄9∶1组、茵黄1∶9组ALT水平降低;茵黄3∶1组AST、茵黄10∶0组,茵黄3∶2组、茵黄1∶1组、茵黄1∶9组、茵黄0∶10组AST水平降低,差异均有统计学意义(P0.05)。与模型组比较,茵黄3∶2组、茵黄1∶1组、茵黄1∶9组、茵黄0∶10组TC水平下降,茵黄1∶9组、茵黄0∶10组TG水平下降,差异均有统计学意义(P0.05)。病理学形态分析结果显示,各治疗组均能明显改善肝组织损伤的程度。结论:茵黄不同比例治疗组可抑制肝损伤小鼠血清中ALT、AST升高的趋势,对四氯化碳急性肝损小鼠伤均有保护作用,以茵黄配比1∶9组效果最佳。

Abstract:Objective:To compare the protective effects of combination of Yinc hen(Herba Artemisiae Scopariae) and Dahuang (Radix et Rhizoma Rhei) (Yin Huang combination) of different proportions on mice liver injury induced by CCl and explore the optimal proportion of Yinchen (Herba Artemisiae Scopariae) and Dahuang (Radix et Rhizoma Rhei) for the mice liver injury induced by CCl.Methods:Totally 80 mice were randomized into blank group,model group,positive control group and Yin Huang (10 ∶ 0) group,Yin Huang (9 ∶ 1) group,Yin Huang (3 ∶ 1) group,Yin Huang (3 ∶ 2) group,Yin Huang (1 ∶ 1) group,Yin Huang (1 ∶ 9) group,Yin Huang (0 ∶ 10) group,8 mice in each group.Positive control group were performed gavage of Bifendate (0.2 g · kg-1) while treatment group were given gavage of Yin Huang combination of the above proportions.Blank group and model group were given gavage with distilled water of same volume.Each group was given continuous gastral administration of 7 days.One hour after the last administration,administration groups and model group were given single abdominal injection of carbon tetrachloride peanut oil solution (0.01 mL · g-1) with mass score of 0.1%.After 16h,the blood was taken by removing eyeballs,and the levels of ALT and AST were measured.The weight of liver was determined by taking out the liver and the hepatosomatic index was calculated.Liver tissue bomogenate was made and the concentrations of TG and TC were determined.Tissue sections were made bv the same part of the liver,and the pathologic morphology was observed after HE staining.Results:Compared with model group,Yin Huang combination with different proportions can reduce the hepatosomatic index in mice with liver injury;the difference had statistical significance (P < 0.05).Combined with blank group,the levels of ALT and AST increased markedly;compared with model group,levels of ALT in Yin Huang (10 ∶ 0) group,Yin Huang (9 ∶ 1) group and Yin Huang (1 ∶ 9) group declined;levels of AST in Yin Huang (10 ∶ 0) group,Yin Huang (3 ∶ 2) group,Yin Huang (1 ∶ 1) and Yin Huang (0 ∶ 10) group declined,with statistical difference (P < 0.05).Compared with model group,levels of TC in Yin Huang (3 ∶ 2)groulp,Yin Huang (1 ∶ 1) group,Yin Huang (1 ∶ 9) and Yin Huang (0 ∶ 10) group declined,with statistical difference (P <0.05).The results of pathologic morpbology,each treatment group can obviously improve the degree of liver tissue injury.Conclusion:Yin Huang combinations of different proportions can inhibit the increase of levels of ALT and AST in the serum of mice with liver injury,which has protective action for the liver injury induced by CCl and the optimal proportion is Yin Huang (1 ∶ 9).
