恙螨RAPD-PCR初步研究

摘    要:通过制备恙螨的基因组DNA,利用一组10个碱基序列的单链随机引物,建立恙螨的随机引物DNA多态性分析技术(RAPD-PCR),为恙螨的种间、种内、种下、种群之间的分子分类、遗传特性、亲缘关系等研究打基础。本研究筛选出N01,N05,N08,N104个随机引物,较适的缓冲液pH值,Mg ̄(2+)浓度和模板DNA浓度,以及PCR实验参数。

Abstract:Larvae of chigger mites are the vectors of scrub typhus.There are still many problems and different opinions on differentiating and identifying the chigger mites. In this study.after extracted the genomic DNA from chigger mites,with 10-mer single-stranded DNA primers,the approaches to RAPD-PCR in chigger mites were established.This technique can be used to differentiate and identify the species groups,cryptic species,sibling species,interspecies,intraspecies,subspecies,population,genetic characteristics of chigger mites and their relationship.The primers of N01,N05,N08,N10 were selected in this experiment, the suitable pH(8.3),Mg2+concentration(about2.0mmol/L),template DNA concentration and the parameters of PCR were settled.
