二甲亚砜对人巨细胞病毒初次分离促进作用的实验观察

目的观察二甲亚砜(DMSO)对人巨细胞病毒(HCMV)初次分离的影响.方法将临床收集的11例患者尿标本接种至两组人胚成纤维细胞(HF)细胞中进行HCMV分离.1组为DMSO细胞组,即在培养液中添加了终浓度为1%的DMSO;另1组为常规细胞组接种组,设为对照,不作任何处理.待细胞出现HCMV特征性细胞病变(CPE)时,通过PCR法测定HCMV DNA加以确证.结果 11例标本中,两组均有5例标本出现HCMV CPE,且标本号一致.但DMSO细胞组CPE出现时间比对照组提前约9 ～12 d.结论 DMSO对HCMV初次分离的阳性结果没影响,但可使HCMV特征性CPE的出现得以提前,加快了临床标本中HCMV的分离.

Effect of DMSO on the first isolation of human cytomegalovirus

Objective To evaluate the effect of DMSO on the isolation of human cytomegalovirus. Methods Eleven urine specimens from clinical kidney transplant recipients and children of cerebral palsy, were inoculated into two groups of human fibroblast cells for HCMV isolation. The growth medium of one group contained a final concentration of 1% DMSO; another group was used as control. PCR was used to evaluate the efficacy of isolation. Results Among 11 samples, CPE appeared in two groups, and the samples was unanimous , but the time of CPE appearance was different. The time of the DMSO group was shorter than that of the untreated group obviously. Conclusion DMSO don't enhance the rate of detection of HCMV in shell vial cell cultures , but have significant effect on the time of CPE appearance compared with that in untreated cultures.
