6-姜酚对脑缺血再灌注后SGK1和FAS表达的影响

摘    要:目的探讨6-姜酚对大鼠局灶性脑缺血再灌注损伤后SGK1表达的影响。方法采用线拴法建立大鼠大脑中动脉栓塞(MCAO)模型。大鼠随机分为缺血组、假手术组、6-姜酚小剂量组(2mg/kg)、6-姜酚大剂量组(4mg/kg)。缺血2h,再灌注24h后处死动物,于缺血前30min和再灌注前30min舌下静脉给药。采用排水法测定脑梗死体积,采用Western blot方法检测脑组织SGK1和凋亡相关基因FAS的表达。结果6-姜酚治疗组与模型组相比脑梗死体积缩小,脑组织SGK1表达上调,凋亡相关基因FAS表达减少。结论6-姜酚对大鼠局灶性脑缺血再灌注损伤具有保护作用,其作用机制可能与增加SGK1表达及抑制凋亡相关基因FAS的表达有关。

Effect of 6-ginger on the Expression of SGK1 and FAS in Rats with Focal Ischemia-reperfusion Injury

Abstract:Objective To investigate the effect of 6-ginger on the expression of SGK1 and FAS in rats with middle cerebral artery occlusion (MCAO). Methods Male Sprague-Dawley rats were anesthetized and subjected to 2h MCAO followed by 24h reperfusion. The neurological deficits and cerebral infarction volume were measured. Western blot was used to detect the expression of SGK1 and FAS protein. Results The neurological deficits and cerebral infarction volume were decreased in 6-gingerol-treated groups compared with sham opera- tion group. The expression of SGK1 in 6-gingerol-treated group was higher than that of ischemic group. 6-gingerol-treatment also decreased the expression of FAS protein. Conclusion 6-gingerol has protective effect on focal cerebral ischemic reperfusion injury through inhibiting FAS expression and upregulating SGK1 expression.
