骨膦抗去势雌鼠骨质疏松研究

摘    要:切除8月龄雌性大鼠双侧卵巢后,皮下注射骨膦12周,与单纯卵巢切除组(OVX组)、假手术组(sham组)及口服尼尔雌醇组(E3组)比较,用骨生物力学、骨密度测量、骨形态计量学等方法对骨膦的药效进行综合评价。结果表明,与sham组比较,OVX组骨灰重、骨干重、骨钙含量及骨密度、抗弯力、骨小梁体积密度、宽度均明显降低,而成骨细胞指数和破骨细胞指数、尿羟脯氨酸则明显增高。骨膦和E3对上述指标有显著改善作用。

Experimental study on anti-osteoporosis effect of clodronate in ovariectomized female Rats

Abstract:In order to evaluate the anti-osteoporosis effect of clodronate in ovariectomized female rats,44 female Wistar rats aged 8 months were divided randomly into 4 groups:a) simple ovariectomized the (OVX)group; b)sham-operated group ;c)the E3 group which were subjected to bilateral ovariectomy and treated on the next day with nilestriol ling/kg BW by gavage once a week for 12 weeks,and d)the clodronate group which were subjeded to bilateral ovariectomy and treated on the next day with clodronate sing/kg BW by subcutaneous injection twice a week for 12 weeks. All the animals were sacrified 12 weeks after the operation. The blood and urine samples were collected for determination of biochemical indices. The humeri and tibiae were prepared for determination of net weight,ash weight,calcium and phosphorus content,bone density,as well as bone histopathology and histomorphometry. The left femora were processed for bone anti-curve power. The results showed that as compared with the sham group,the net weight,ash weight,calcium content,density and bending resistance of bones,the trabecular bone volume and thickness were all significantly reduced in the OVX group,while its osteoblast index,osteoclast index and urine hydroxyproline content were obviously increased. In the E, group and the clodronate group,however,these indices were close to normal levels. The data indicated that clodronate and E3 can all effectively prevent bone loss in ovariectomized female rats, and it seems that the effect of clodronate is stronger than that of nilestriol.
