TRPM7与风湿性心脏病心房颤动患者心房纤维化的关系

目的:检测风湿性心脏病心房颤动患者心房组织中瞬时受体电位通道亚族M7(TRPM7)的mRNA及蛋白水平,探讨TRPM7与房颤心房纤维化的关系.方法:19例风湿性心脏病患者于换瓣术中获取约200 mg的右心房肌组织,分为窦律(SR)组9例和房颤(AF)组10例.HE病理染色观察心肌细胞横截面积,Masson染色及心肌纤维化标志物观察心房纤维化,Western blotting和实时定量PCR测定心房肌组织中TRPM7的mRNA及蛋白水平.结果:与SR组比较,心肌细胞横截面积、胶原容积分数、心肌纤维化标志物CTGF、Collagen Ⅰ和CollagenⅢ的mRNA水平、TRPM7的mRNA及蛋白水平在AF组显著增加,差异具有统计学意义(P＜0.05).结论:风湿性心脏病房颤患者心肌组织中TRPM7的表达增加,可能通过促心房纤维化,参与房颤的发生与维持.

Relationship of TRPM7 and Atrial Fibrosis in Rheumatic Heart Disease Patients with Atrial Fibrillation

Objective: To investigate the possible relationship between transient receptor potential melastatin 7(TRPM7) and atrial fibrosis in rheumatic heart disease patients with atrial fibrillation.Methods: About 200 mg right atrial arterial tissue sample was extracted from each of the 19 patients with rheumatic heart disease,including 9 cases with sinus rhythm(SR) and 10 cases of atrial fibrillation(AF),during valve replacement surgery.HE,Masson stained sections and myocardial fibrosis markers(CTGF,collagen Ⅰ,and collagen Ⅲ) were used to determine the cross-sectional area and level of fibrosis respectively.The mRNA and protein levels of TRPM7 in atrial tissue were measured by Western blotting and real-time quantitative PCR.Results: Compared with SR group,the cross-sectional area of the myocardial cells,collagen volume fraction,the mRNA levels of myocardial fibrosis markers such as CTGF,collagen Ⅰ and collagen Ⅲ,the mRNA and protein levels of TRPM7 in the AF group were significantly increased(P<0.05).Conclusion: The expression of TRPM7 increased in the myocardial tissue of patients with rheumatic heart disease which may promote atrial fibrosis,and then involved in the occurrence and maintaining of atrial fibrillation.
