氨甲环酸对中波紫外线照射后黑素细胞影响的研究

摘    要:目的:探讨氨甲环酸对中波紫外线照射后黑素细胞增殖及酪氨酸酶代谢的影响。方法:设立紫外线照射组和非照射组,以不同浓度的氨甲环酸作用于黑素细胞,cck-8法测定黑素细胞增殖;多巴氧化方法测定酪氨酸酶活性,RT-PCR方法测定酪氨酸酶mRNA表达水平。结果:氨甲环酸对两组黑素细胞的增殖均无明显抑制作用(P0.05);而对酪氨酸酶活性和酪氨酸酶的mRNA表达有抑制作用(P0.05),两组结果比较无明显差异;结论:氨甲环酸对黑素细胞的酪氨酸酶的抑制作用与中波紫外线照射无直接关系。

Abstract:Objective To study the impact of tranexamic acid on cell proliferation and tyrosinase activity in cultured human melanocytes after ultraviolet irradiation. Methods To set up the ultraviolet irradiation and irradiation group, melanocytes was incubated different concentrations of tranexamic acid.Cell proliferation was measured with cck-8 assay;The tyrosinase activity was detected by DOPA-oxidation;Tyrosinase mRNA level was detected by RT-PCR method. Results Melanocytes proliferation was not significantly inhibitted by tranexamic acid compared with the untreated control (P〉0.05);but tyrosinase activity and tyrosinase mRNA level was restrained respectively (P〈0.05).The conclution was similary. Conclusion The tyrosinase activity was directly inhibitted by tranexamic acid,thereby melanogenesis was reduced,This result is not affected by ultraviolet irradiation.
