硝苯啶治疗消化性溃疡的疗效及其机理探讨

用硝苯碇预防无水乙醇和幽门结扎所致大鼠胃溃疡，结果显示用药组胃溃疡指明显低于对照组，对溃疡的抑制率与ＮＦＤ剂量呈正相关。临床观察４２例十二指肠球部溃疡患者，服用ＮＦＤ １０ｍｇ，每日４次，６周后溃疡愈合２７例，总有效３９例，与对照比较有显著差异。

An Observation of Nifedipine Effect on the Treatment of Peptic Ulcer and a Study on its Mechanism

It was observed that nifedipine(NFD), given intraperitoneally, protected the gastric mucosa of rats from gastric ulcer induced by gavage with absolute ethanol and pyloric ligation. The gastric ulcer inhibition percentage is positively related to the dosage of NFD. 42 duodenal ulcer patients diagnosed with pastrocopy were treated with 10 mg NFD qid for 6 weeks. 27 cases of the 42 healed(64. 3%). The total effective rate was 92.9% . There is a significant difference between NFD group and the control. About 20 gastric ulcer cases were treated as the same way. 15 patients healed(75.0%). The total effective rate was 95. 0%. The study on the antiulcer mechanism of NFD demonstrated that NFD reduced the volume of gastric liquid, gastric acid secretion and pepsin activity. There are significant differences between NFD group and the control ( P<0.05). Measured by radioimrnunoassay at 1. 5 and 2 hrs after absolute ethanol gavage respectively, the values of prostaglandin E2(PGE2) synthesis in NFD 10 mg group were higher than that in control (P<0.05 and P<0.01, respectively). NFD's effect on treating peptic ulcer is similar to that of cimetidine and with small side effect. It does not affect the blood pressure of a patient with normal blood pressure. It is cheap and convenient. Therefore, it can be used as a new drug in the treatment of peptic ulcer.
