庆大霉素致大鼠肾近端小管上皮细胞凋亡的超微结构研究

摘    要:目的探讨庆大霉素对实验性大鼠肾近端小管上皮细胞凋亡的影响。方法通过给大鼠腹腔注射庆大霉素100mg/Kg/d,分别在连续用药第5天、8天、10天等不同天数处死大鼠,取肾脏制备超薄切片,在透射电镜下观察肾近端小管上皮细胞凋亡的情况。结果大鼠连续注射庆大霉素5天、8天、10天后肾近端小管上皮细胞均有凋亡的发生,注射庆大霉素10天的大鼠肾小管上皮细胞出现较多坏死。结论庆大霉素肾毒性作用部分是通过肾近端小管上皮细胞凋亡而实现的,细胞凋亡的发生与庆大霉素肾毒性作用强弱有关。

Abstract:Objective To discuss the effects of gentamicin nephrotoxicity on the apoptosis of proximal tubule epithelial cells of rats. Methods Gentamicin was injected to rats intraperitoneally 100mg/Kg/d, and the rats were killed at the 5th, 8th, and 10th day respectively after continuous drug injection. The nephridal tissue of the rats was collected and was made into ultrathin sections. The apoptosis of proximal tubule were observed by the use of transmission electron microscope. Results Apoptosis could be seen at the 5th, 8th, and 10th day after continuous gentamicin injection, and at the 10th day, necrosis could also been seen. Conclusion Gentamicin nephrotoxicity was implemented through apoptosis of proximal tubule epithelial cells, and the onset of apoptosis was related to the intensity of gentamicin toxicity.
