杜仲诱导大鼠bMSCs分化早期成骨/成脂相关基因表达变化

摘    要:目的:观察杜仲醇提取物诱导骨髓间充质干细胞(bMSCs)分化早期成骨/成脂相关基因表达变化。方法:培养大鼠bMSCs,用杜仲醇提取物分别诱导bMSCs 8 h,1、3、7 d,采用荧光定量PCR(RT-qFCR)的方法检测成骨相关基因Id4、Runx2、ALP和成脂相关基因PPARγ、Adipoq、aP2的表达变化。结果:Id4诱导3 d后表达略有上调;Runx2基本无变化;ALP诱导7 d后显著升高;PPARγ和aP2诱导3 d达最高峰,然后下调;Adipoq诱导7 d显著上调。结论:杜仲对bMSCs的成骨/成脂分化具有双重调节作用,提示杜仲醇提取物中既含有促成骨分化,也含有促成脂分化的成分。

Abstract:Objective: To observe the expression of osteogenic and adipogenic differentiation related genes at early period in rat bone mesenchymal stem cells(bMSCs) induced by eucommia bark.Methods: bMSCs were incubated with MeOH extract of eucommia bark for 8 h,1 d,3 d,and 7 d.Realtime PCR was used to detect the expression of osteogenic related genes,Id4、Runx2 and ALP,and adipogenic related genes,PPARγ、Adipoq and aP2 compared to control groups.Results: Id4 upregulated slightly after induction 3 days later,Runx2 did not changed basically,ALP increased obviously after induced for 7 days,PPARγ and aP2 reached the highest point at 3 days and then downregulated and Adipoq increased obviously after induced for 7 days.Conclusion: Eucommia bark may regulate the expression of genes both osteogenic and adipogenic on bMSCs,and this implies that eucommia bark contains compositions which can promote osteoblastogenesis and adipogenesis.
