不同时机介入电针对模拟失重大鼠肝脏HSP 70、MDA、SOD和GSH-PX的影响

摘    要:目的:观察预针刺和针刺治疗对模拟失重大鼠肝脏超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)活力,丙二醛(MDA)含量及热休克蛋白70(HSP 70)表达的影响,探讨电针对模拟失重大鼠肝脏损伤改善作用的相关机制。方法:Wistar大鼠随机分为正常组、模型组、预针刺组和针刺组,每组5只。大鼠尾部悬吊4周复制模拟失重模型。预针刺组在尾部悬吊前1周,电针刺激双侧"肾俞""脾俞"和"三阴交"穴,每次30min,每日1次;针刺组大鼠尾部悬吊过程中电针刺激"肾俞""脾俞"和"三阴交"穴,每次30min,隔日治疗1次,共针刺14次。采用免疫组化法检测大鼠肝脏组织HSP 70的表达情况,比色法检测大鼠肝脏组织SOD、GSH-PX活性和MDA含量。结果:与正常组比较,模型组大鼠肝脏HSP 70呈强阳性反应,HSP 70表达量显著增加(P0.01),MDA含量显著升高(P0.01),SOD和GSH-PX活性显著降低(P0.05);与模型组比较,预针刺组大鼠肝脏HSP 70表达量显著降低(P0.01),SOD和GSH-PX活性略有升高(P0.05),MDA含量略有减少(P0.05),针刺组HSP 70表达量略有降低(P0.05),SOD活性和MDA含量略有升高(P0.05),GSH-PX活性略有降低(P0.05);与预针刺组相比,针刺组肝脏GSH-PX活性显著降低(P0.05),MDA含量显著升高(P0.05)。结论:预针刺可以明显抑制大鼠尾部悬吊引起的肝HSP 70表达的上调,从而可能有利于提高肝脏的抗氧化能力。

Effect of Electroacupuncture Intervention at Different Time-points on Levels of HSP 70,MDA,SOD and GSH-PX of Liver in Rats with Simulated Weightlessness

Abstract:Objective To observe the effect of acupuncture intervention at different time-points on the activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-PX),the content of malonaldehyde(MDA)and expression of heat shock protein 70(HSP 70)of liver tissue in rats with simulated weightlessness,so as to explore its mechanism underlying improvement of liver injury in rats with simulated weightlessness.Methods Twenty male Wistar rats were randomly divided into control group,model group,pre-acupuncture group and EA group,5rats in each.The model of simulated weightlessness was established by tail suspension for 4week.One week before the tail suspension,the rats in the pre-acupuncture group were treated with electroacupuncture(EA)at"Shenshu"(BL 23),"Pishu"(BL 20)and "Sanyinjiao"(SP 6)for 30 min before treatment,once a day for 7days.The rats in the EA group received tail suspension and acupuncture intervention at the same time.EA was applied for 30 min per treatment,once every other day for 14 times.Immunohistochemical staining was used to assay the expression of HSP 70 in the liver tissue.The activities of SOD and GSH-PX and content of MDA in liver tissues were examined by means of colourimetric method.Results Compared with the control group,the expression of HSP 70 and the content of MDA in the liver tissue were increased significantly(P<0.01),and the activity of SOD and GSH-PX was notably reduced(P<0.05)in the model group.Compared with the model group,the content of HSP 70 was significantly reduced in the pre-acupuncture group(P<0.01).There were no significant changes in the levels of SOD,GSH-PX,MDA and HSP 70 in the EA group(P>0.05).In comparison with the pre-acupuncture group,the activity of GSH-PX was lower(P<0.05)and the content of MDA was higher(P<0.05)in the EA group.Conclusion EA-pretreatment can suppress the increase of liver HSP 70 immunoactivity in rats with simulated weightlessness,being likely to improve the antioxidant ability of liver.
