我国恶性疟原虫地理株SSUrDNA特定序列的变异分析

摘    要:采用聚合酶链式反应(PCR)技术,分别从我国安徽、海南及云南三省疟区来源的恶性疟原虫株基因组DNA中,扩增出该虫小亚单位核糖体核糖核酸(SSUrRNA)基因特定片段,长约570bp,将其插入M13mp18和M13mp19载体中,以双脱氧末端终止法测序。读序结果分析表明,恶性疟原虫地理株间及与巴西IMTM22株7G8克隆间,SSUrDNA特定片段序列具有高度同源性,差异仅表现为个别碱基置换、插入或缺失所致的点突变,且均发生在可变区中,其发生频率亦无明显差别。

Abstract:In this paper,specific SSUrDNA fragments about 570 bp from genomic DNA re-spectively extracted from Plasmodium falciparum isolates of Anhui, Hainan and Yunnanprovinces in China were amplified with polymerase chain reaction.The amplified DNA fragmentsof P.falciparum isolates from three provinces were inserted into vector M13 mp18 and mp19,then the sequences were determined with dideoxynucleotide terminator method and compared eachother by computer programming.The results showed that there was high homology amongstSSUrDNA sequences of P.falciparum isolates including 7G8 clone reported outside China withvery fewer point mutation resulting from nucleotide substitution and deletion or insertion whichoccured in variable region of SSUrDNA,and no significant diversity in the rate of point mutationsof SSUrDNA sequences from different geographical isolates of P.falciparum was noticed.
