OYX-OP大鼠腺垂体FS细胞中S-100蛋白表达的共聚焦显微镜研究

摘    要:目的:探讨神经组织蛋白S-100在去卵巢致骨质疏松症大鼠(OVX-0P)腺垂体滤泡星形细胞(FS细胞)中的表达。方法:采用适龄SD大鼠40只,随机均分为卵巢切除(OVX)组和假性手术对照(Sham组),于术前和术后6周末测量大鼠全身及腰椎骨密度(BMD)。取垂体,应用免疫荧光组织化学法结合激光扫描共聚焦显微镜(LSCM)分析技术,检测FS细胞中S-100表达。结果:术后6周末OVX组大鼠全身及腰椎BMD均明显低于Sham组值(P<0.01,P<0.01)。S-100阳性荧光标记物定位于FS细胞的胞浆中。FS细胞具有典型的细胞突结构、与腺垂体中丰富的血窦直接相邻、结成了腺垂体中发达的活性细胞网络、在结构上与多种腺垂体内分泌细胞密切相连。两组间S-100荧光强度和表达阳性率无显(P>0.05)。结论:SD大鼠OVX术后6周出现骨质疏松变化,FS细胞在形态结构和定位上的特征及其在数量上的稳定性可能是中枢调节骨代谢因素作用的一个细胞基础。

Abstract:Objective To investigate the expression of nerve tissue protein S-100(S-100)in adenohypophyseal folliculo stellate cells(FSCs) in ovariectomized osteoporotic rats. Methods Osteoporotic rat models were obtained by bilateral ovariectomy. 40 female rats were equally randomized into ovariectomized group(OVX group)and sham operatied control group(sham group).Total skeletal and spinal bone mineral density(BMD) of each rat were measured by DEXA.The expression of S-100 in adenohypophyseal glands was detected by immunofluorescent histochemistry using laser scanning confocal microscopy(LSCM). Results Total skeletal BMD and spinal BMD of rats in OVX group were significantly lower than those in sham group( P <0 01, P <0 01).S-100-positive FSCs in anterior pituitary(AP)were indentified by their unique features:cytoplasmic processes between the endocrine cells and adjacent to blood sinuses.There was no difference in fluorescence and positive rate of S-100 in the cytoplasms of FSCs between OVX group and sham group( P >0 05). Conclusion Osteoporosis in OVX rats can be determined by significantly decreased BMDs.The microscobic characteristics and quantitative identity of FSCs in either group revealed by detecting S-100 could presumably form a cellular basis of bone metabolism in AP.;
