针刺对大鼠颅脑损伤后Nogo-A表达的实验研究

摘    要:[目的]观察Nogo—A在大鼠颅脑损伤后表达变化,探讨针刺干预颅脑损伤后大鼠Nogo—A表达的影响。[方法]雄性SD大鼠随机分为针刺治疗组、模型对照组、正常对照组,用改良Feeney’s打击法制成大鼠左顶叶局限性脑挫裂伤模型,分别于损伤后6、24、48h和6d组取损伤局部脑组织,采用免疫组化法观察Nogo-A的表达变化并进行统计学分析。[结果]颅脑损伤后,从24h开始,针刺组大鼠脑损伤组织中Nogo—A的表达开始下降。与模型组相比较,针刺组48h、6d的Nogo—A表达均显著降低,差异有统计学意义(P〈0.01)。[结论]针刺治疗对大鼠颅脑损伤后Nogo—A表达具有明显的抑制作用,表明针刺对脑损伤大鼠的神经再生具有促进作用。

Abstract:[Objective] To observe variation of Nogo-A expression after craniocerebral injury(CCI)of rats,and to investigate the protective mechanism of acupuncture in CCI. [Methods] Rats were randomly divided into three groups: acupuncture group, model group and blank control group. The craniocerebral injury rat model was made by using a modified Allen's method. The craniocerebral tissues on 6, 24, 48 h and 6 d after injury were taken. The expression of Nogo-A was detected through immunohistochemistry mothods. The positive cells were analyzed by a computer image analysis system. [Results] After CCI rat model was established successfully,and the Nogo-A content of local injury tissues in acupuncture group began to decline from 24 h. Compared with the model group,statistical significance was found in the acupuncture group at 48 h, 6 d(P<0.01).[Conclusion] The acupuncture can obviously inhibit the expression of Nogo-A of the CCI rat and it is likely to play a role in the axon regeneration along the correct direction after CCI.
