局灶性脑缺血大鼠白细胞流变性变化及乌龙丹的影响作用

摘    要:背景 缺血性脑血管病白细胞流变性与微循环密切相关,传统活血化瘀中药是否对白细胞流变学异常、变形能力下降有影响?目的通过建立大鼠局灶性脑缺血病理模型,观察其白细胞流变性的变化,并探讨活血化瘀中药复方乌龙丹的作用.设计随机对照的实验研究. 地点和材料实验地点解放军第一军医大学中医系实验室.选用Wistar雄性大鼠 40只,体质量( 250± 20) g,随机分为假手术组、模型组、乌龙丹低剂量组、乌龙丹高剂量组,每组10只. 干预大鼠结扎颈总动脉( CCA)后再阻断大脑中动脉( MCA)造模,造模术后24 h取血用核孔滤膜红细胞变形能力测定仪测定白细胞变形指数、白细胞滤过堵塞的粒子浓度及红、白细胞-红细胞滤过指数.主要观察指标实验各组大鼠白细胞总数、白细胞变形指数、白细胞滤过堵塞的粒子浓度及红、白细胞-红细胞滤过指数.结果模型组的白细胞总数、白细胞变形指数、白细胞滤过堵塞的粒子浓度及红、白细胞-红细胞滤过指数均明显高于假手术组(P< 0.01),表示白细胞的变形能力下降.乌龙丹给药组上述指标与模型组有显著性差异(F=3.761, P< 0.01),显示出该方对梗死后白细胞变形能力下降有一定的对抗作用.结论 急性局灶性脑缺血损伤中有白细胞流变学异常,变形能力下降.中药复方乌龙丹对此有一定对抗作用,增强白细胞的流变性和使其黏附性降低,是其活血化瘀作用的重要方面.

Abstract:BACKGROUND:Leukocyte rheology in ischemic cerebral vascular disease is closely related to microcirculation.Whether do the traditional Chinese herbs for activating blood circulation and removing stasis reduce the abnormal leukocyte rheologic change and deformability? OBJECTIVE:To probe into the action of Chinese herbal compound wulong dan on activating blood circulation and removing stasis so as to observe leukocyte rheologic change by establishing the pathological model of focal cerebral ischemia in rats. DESIGN:A randomized and cotrolled trial design. SETTING and MATERIALS:The experiment was performed in the Laboratory of Department of Traditional Chinese Medicine in First Military Medical University of Chinese PLA.Forty Wistar male rats,weighing(250± 20) g,were randomly divided into sham-operation group,model group,low-dosage group and high-dosage group,10 rats in each group in the experiment. INTERVENTIONS: The model was prepared by blocking middle cerebral artery after the common carotid artery ligature.Twenty-four hours later,blood was collected to measure the ameboid index of leukocytes,the percentage of the particle blocked by the filtration of leukocytes,filtration indexes of erythrocyte and leukocyte-erythrocyte with nucleopore filter membrane erythrocyte deformability meter. MAIN OUTCOME MEASURES:The total count of leukocytes,deformity index of leukocytes,the percentage of the particle blocked by the filtration of leukocytes,filtration index of erythrocytes and leukocyte-erythrocyte of rats in each group in the experiment. RESULTS:In model group, the total count of leukocytes, deformity index of leukocytes,the percentage of the particle blocked by the filtration of leukocytes,filtration index of erythrocytes and leukocyte-erythrocyte were remarkably higher than those in sham-operation group(P< 0.01),indicating that the deformability of leukocytes was declined.In wulong dan group, there were significant differences concerning to the above-mentioned indexes,compared with model group(F=3.761,P< 0.01), indicating the formula served a certain action of antagonism to the decline of deformability of leukocytes after infarction. CONCLUSION:The acute focal cerebral ischemic injury leads to the abnormal leukocyte rheology and declining of deformability.The Chinese herbal compound wulong dan serves a certain action of antagonism, enhances rheologic property of leukocyted and reduces adhesion,which is taken as the essential part for its action of activating blood circulation and removing stasis.
