国产红葡萄酒对食饵性动脉粥样硬化病变的消退作用及其机制的研究

摘    要:目的通过复制8周兔的早、中期动脉粥样硬化(AS)模型,探讨红葡萄酒(RW)对8周AS兔病变的逆转作用及其可能的机制。方法采用HE染色、电泳流动漂移技术(EMSA)、免疫组织化学和原位杂交技术检测食饵性动脉粥样硬化8周时血管壁病理形态学改变和核转录因子(NF-κB)活化及单核细胞趋化蛋白(MCP-1)表达情况及RW再干预8周后对AS血管壁病理形态学及对NF-κB活化和MCP-1表达的影响。结果RW治疗AS 8周后,其主动脉硬化斑块面积、内膜增生厚度、泡沫细胞数及平滑肌增生程度均有明显减轻;在AS斑块明显缩小的前提下,其MCP-1的蛋白及其mRNA水平的表达均有所下降,且NF-κB活性受到明显抑制。结论RW对AS病变具有一定的消退作用,其机制可能系通过抑制NF-κB活化,及下调MCP-1的表达而实现的。

Abstract:Objective To study the therapeutic effect of red wine(RW) made in china on experimental atherosclerosis.Methods Eight-weeks experimental atherosclerosis(AS) model was set for studying the effect of red wine at cellular,molecular and gene levels.Light microscopy,electrophoretic mobility shift assay(EMSA),in situ hybridization and immunohistochemistry were used to study morphological character,activity of NF-kB and the expression of monocyte chemotactic protein-1(MCP-1) in atherosclerotic plaques at 8 weeks of atherosclerosis and after treatment for 8 weeks.Resultes After treatment with RW for 8 weeks,RW group had lighter aortic AS,less lipid infiltration and smooth muscle cell proliferation,decreased thickness of tunica intima and elastic fibers,etc,in the plaques compared with those in AS group,RW could remit the formed AS plaque by suppressing the NF-κB activity and down-regulating the expression of MCP-1.Conclusion The results indicated that RW had the therapeutic effect on AS and reduced its formation and progression by suppressing the NF-κB activity and down-regulating the expression of MCP-1.
