不同干燥方法对祛湿化瘀方 防治实验性脂肪肝药效学的影响

摘    要:目的:探讨不同干燥方法对"祛湿化瘀方"防治实验性脂肪肝药效学的影响。方法:祛湿化瘀方中5味药物分别醇提、水提醇沉后合并,制备成生药含量为0.93 g.mL-1的药液,再按不同干燥方法制得浸膏、70℃真空负压干燥药物、40℃真空负压干燥药物、冷冻干燥药物、喷雾干燥药物。采用Wistar雄性大鼠,以四氯化碳(CCl4)复合高脂低蛋白饮食制备大鼠脂肪肝模型,造模2周后,将造模大鼠随机分为模型组、未干燥组、浸膏组、70℃真空负压干燥组、40℃真空负压干燥组、冷冻干燥组、喷雾干燥组,每组灌以相应药物2周。取材后检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、谷氨酰转移酶(GGT)活性、肝组织甘油三酯(TG)、游离脂肪酸(FFA)的含量变化。并对各个药物样品进行指纹图谱分析。结果:与正常组相比,模型组ALT,AST,GGT活性,肝组织TG,FFA含量显著升高。未干燥组与模型组相比,血清ALT,AST,GGT及肝组织TG,FFA显著下降;浸膏组与模型组相比,血清GGT,ALT,肝组织TG,FFA显著下降;70℃和40℃真空干燥组除血清ALT活性显著降低外,其余指标与模型组相比均无显著差异;冷冻干燥组除肝组织TG显著低于模型组,其余指标与模型组相比均无显著差异;喷雾干燥组除血清GGT显著低于模型组,其余指标与模型组相比均无显著差异;未干燥组的综合疗效优于其他各组。运用指纹图谱对各组药物进行分析,色谱图有相应的谱峰信息综合表达有差异。结论:"袪湿化瘀方"经不同干燥方法后对实验性脂肪肝药效均有所下降,不同方法干燥后部分有效物质丢失,可能是导致药效差异的主要原因。
