吴茱萸致小鼠肝毒性时效、 量效关系研究

摘    要:目的:研究吴茱萸致小鼠肝毒性的时效、量效关系。方法:将KM小鼠随机分为空白组和吴茱萸组,吴茱萸组分为5个不同时间组和3个不同剂量组。不同时间组动物在连续ig吴茱萸水煎液50 g.kg-1后于8,15,20,25,30 d处理,不同剂量组以100,50,25 g.kg-1连续ig 8 d后处理,各组动物均检测血清丙氨酸氨基转移酶(ALT),天冬氨酸氨基转移酶(AST),计算肝脏指数,并观察肝组织病理学变化。结果:与正常组相比,吴茱萸50 g.kg-1能够升高小鼠ALT,AST水平、肝脏指数(P<0.01),且ALT,AST水平在给药后第20 d达高峰,ALT从(31.7±9.1)U.L-1升高至(621.4±244.9)U.L-1,AST从(120.0±31.8)U.L-1升高至(407.6±132.8)U.L-1,肝脏指数和病理形态学的改变也具有时间依赖性;给予不同剂量的吴茱萸后,小鼠血清ALT,AST水平、肝脏指数均有上升,中高剂量组ALT分别为(127.2±42.0),(320.0±131.3)U.L-1,AST分别为(282.1±69.8),(471.4±143.7)U.L-1,与正常组相比ALT,AST水平和肝脏指数差异非常显著,病理形态学改变随剂量增大而损伤严重,毒性呈剂量依赖性。结论:小鼠连续ig给予吴茱萸水煎液可以造成肝脏的损伤,毒性与时间、剂量呈一定相关性。
