血府逐瘀胶囊干预SIRT1通路介导缺血细胞形态学变化的研究

目的观察血府逐瘀胶囊对大鼠缺血心肌超微结构的改变,从而探讨血府逐瘀胶囊干预沉默信息调节因子2同源蛋白1(Silent mating type information regulation 2 homolog-1,SIRT1)信号转导通路对细胞凋亡形态学的影响,为中医药治疗缺血性心脏病提供依据和新的研究着眼点。方法将80只大鼠随机分为正常组,假手术组,缺血组和血府逐瘀大、中、小剂量组,白藜芦醇组,L-NAME组,给药结束后运用透射电镜观察血府逐瘀大、中、小剂量、白藜芦醇、L-NAME对缺血心肌细胞凋亡形态学的影响。结果各组形态学比较,差异有统计学意义。结论血府逐瘀胶囊可抑制缺血心肌细胞凋亡,其机制可能是通过作为SIRT1抑制细胞凋亡通路的增敏剂而发挥作用。

Research on the Morphological Changes of Ischemic Cell Mediated via SIRT1 Pathway in the Intervention with Xuefu Zhuyu Capsules

Objective To observe the alternation of the ischemic myocardial ultrastructure in the treatment of xuefu zhuyu capsules in the rats and explore the impacts on the apoptosis morphology in the in- tervention of the capsules on the signal transduction pathways of the silent mating type information regulation 2 homolog - 1 （ SIRT1 ） so as to provide the evidences for the treatment of ischemic cardiac disease with Chi- nese medicine and the new research focus. Methods 80 rats were randomized into a normal group, a pseudo - operation group, a model group and the high - dose, middle - dose and low - dose xuefu zhuyu capsules groups, as well as a resveratrol group and a L - NAME group. At the end of medication, the transmission elec- tron microscopy was adopted to observe the impacts on the ischemic myocardiao apoptosis morphology in the high - dose, middle - dose and low - dose xuefu zhuyu capsules groups, as well as a resveratrol group and a L -NAME group. Results The significant differences in morphology were indicated in comparison among groups. Conclusion Xuefu zhuyu capsules inhibit the ischemic myocardial apoptosis, which is achieved probably through the sensitization agent that inhibits SIRT1 apoptosis pathways.
